Tree pony was famous in the world and was originated in China.But its long breeding periodand low propagation coeffidient have always been the big problems for the production andbreeding.Somatic embryogenesis as an improtant approach for plant regeneration and it has beenused successfully for some plants.But currently the research on tree peony were rare.In thisresearch, we used bud scales of Paeonia suffruticosa ‘Fengdanbai’ as explants invitro.Firstly,the effect on embryonic callus induction,POD,SOD,CAT and Soluble Protein contentof pretreating the explants with different Hormones,and histological observe; the effect onembryonic callus induction,POD,SOD,CAT,APX and Soluble Protein content of pretreating theexplants with different Polyamines,and histological observe.Secondly,on basis of the first twotrials, the effect on embryonic callus induction,POD,SOD,CAT,APX and Soluble Protein contentof pretreating the explants with different Hormones and Polyamines,and histological observe.Explore the possible conditions of transformation from non-embryonic callus to embryoniccallus,hoping to provide a theoretical basis for set up a steady regeneration system. The mainresults were as follows:1.Peony callus as material, set different concentration of NAA×6-BA、NAA×TDZ,research the effect on embryonic callus induction of pretreating the explants with Hormones. Theresults showed that: the formation and development of embryogenic callus had something to dowith the species and concentration of Hormones. effect of embryonic callus induction under NAA0.2mg·L-1add6-BA0.5mg·L-1treatment was the best one among the eighteen Hormonestreatment., microscope observe showed that under this condition,embryonic cells formed on20d,on25d appeared a lot of embryonic cells, on25d appeared some early global somatic embryo,non-embryonic and embryonic cells,early global somatic embryo were coexistence. POD activitychanged in the0-20d acute,20-25d increased rapidly and reached the maximum,25-30d declinedslightly,but remained at a high level. SOD activity in0-5d increased rapidly,5-20d remained at ahigh level,20-25d declined,25-30d increased slightly.CAT activity changed in the0-20d acute,20-25d increased rapidly and reached the maximum,25-30d declined. The soluble protein contentchanged slightly in0-10d,10-15d increased and reached peak,15-20d declined thenincreased,reached peak on25d, then declined.2. Peony callus as material, Set different concentration of Put and Spd, research the effect onembryonic callus induction of pretreating the explants with Polyamines. The results showed that: the formation and development of embryogenic callus had something to do with the species andconcentration of Polyamines. effect of embryonic callus induction under Put5mg·L-1treatmentwas the best one among the six Polyamines treatment., microscope observe showed that under thiscondition,embryonic cells formed on25d,on30d appeared a lot of embryonic cells,non-embryonic and embryonic cells were coexistence. POD activity increased then declined in0-10d,10-30d increased and reached the maximum, SOD activity in0-20d showed wavy upwardtrend, reached the maximum on20d then declined. CAT activity in0-25d changed slightly,25-30dincreased. APX activity changed acute, declined then increased in0-15d, reached peak on15dthen declined,25-30d increased. The soluble protein content increased then declined in0-10d,then showed increasing trend,20-25d declined slightly, reached the maximum on30d.3. Peony callus as material, screened two Hormones ratio that suitable for Peony embryoniccallus induction,and on the basis of its,adding different concentrations of Put and Spd respectively.research the effect on embryonic callus induction of pretreating the explants with Hormones andPolyamines. The results showed that: effect of embryonic callus induction was ineffective underthese treatment, not suitable for Peony embryonic callus induction.
