Maize is a most important crop in our country, while in recent years, it has beenseriously endangered by maize rough dwarf disease. The research of this study includes threeaspects: pathogen identification of maize rough dwarf disease in Henan province, the screening ofresistant varieties and pesticide, aims at providing theoretical basis for the prevention and treatmentof maize rough dwarf disease.The main results are as follows:1、The pathogen of maize rough dwarf disease is RBSDV(Rice black-streaked dwarfvirus).The S10ORF coding protein sequences of isolates has a highly consistency with RBSDVS10ORF coding protein sequences on NCBI.Maize samples,which were infected with maize rough dwarf disease, were collected fromHenan province in2012and2013.The isolates were detected with RT-PCR.The analysis of thetarget fragment sequences showed that the RBSDV S10ORF coding protein sequences had agreat consistency of97.0%~97.0%. Except this target fragment, there is no other fragment,which drew a conclusion that the pathogen of maize rough dwarf disease is RBSDV. Theconsistency of RBSDV S10ORF coding protein sequences of2012and2013samples wasanalyzed. At the same time, the phylogenetic tree was built with S10ORF coding proteinsequences of Henan isolates and15RBSDV S10ORF coding protein sequences on GeneBank.From the phylogenetic tree, the evolutionary relationship and genetic relationship of maize roughdwarf disease RBSDV S10ORF coding protein sequences from different areas can be quicklyidentified.2、The screening of resistant varieties shows that most of varieties in our country aresusceptible cultivars and there is seldom resistant cultivar.According to the resistance level of maize decided by the disease incidences,1high resistantcultivars was selected, which was Qinghua686, accounting for1%of all cultivars. And5cultivars,Yuhe863,Wugu305,Dika653, Yudan122, Qinghua696, were moderate resistant with the disease,occupying5%of all cultivars.24samples were resistant with the disease,occupying24%of allcultivars.Moderate susceptible cultivars and high susceptible cultivars occupied70%of all cultivars.It can be seen that most of varieties in our country are susceptible cultivars and there is seldom resistant cultivar, with high resistant cultivars more rare.3、Through the seed coating test,it has been clear that the full fitness gold+sharp4g/ml/kgseeds, imidacloprid high400ml/600g/l+(100kg) of the two groups of agents coating of maizedisease prevention and control effect is better, and can significantly increase the maize yield, so itcan be used as a coating agent for the prevention and control of maize disease.In2013, zhengzhou corn coating agent control effect experiment, result analysis found thatfull fitness gold+sharp4g/ml/kg seed coating of maize disease control effect is best, reach to72.45%, and the corn yield increased by12.02%. In2013, kaifeng qixian corn coating agentscontrol effect experiment, result analysis that found the imidacloprid600grams/liter high400ml/(100kg) coating treatment on maize disease control effect of80.78%, increase maize yield by14.47%.In the year of2013,do the experiments to prevent and control the maize disease withdifferent drug coating in open field test in kaifeng luowang and zhongmou wantan, preventionincrease yield significantly。In2013, kaifeng luownag corn coating agent control effect experiment,result analysis found that rasson seed coating of maize disease control effect is best, reach to79.75%,and the corn yield increased by14.22%. In2013, zhongmou wantan corn coating agentcontrol effect experiment, result analysis found that saiwu seed coating of maize disease controleffect is better, reach to71.61%,and the corn yield increased by11.21%...
