| Gastrodia elata Bl. belonging to Orchidaceae and Gastrodia, which was the Chineserare traditional medicine, is a kind of perennial plant. Tall gastrodia tuber is the drying tuberof Gastrodia elata Bl. It has many kinds of pharmacological action. The modernpharmacology showed that tall gastrodia tuber can improve blood flow, treat withconvulsants, analgesia, and giddy dazzle, increase cerebral blood flow. Gastrodia is the mainactive ingredient in gastrodia elata. The discretion of the gastrodine content in rhizomagastrodiae is an important index to measure the medicinal materials of gastrodia elata blumewhich is good or not. So the China pharmacopoeia showed that the content of gastrodin canevaluate the qualities of gastrodia elata blume. The main raw material is the gastrodia elatafrom hanzhong in this paper. The aim is to study the processing conditions that influenced thecontents of gastrodine in dried fresh Gastrodia elata Bl. and then improve an extraction anddetermination method for gastrodin. On the base of the above work, determine the contents ofgastrodin, gastrodigenin, polysaccharide and the protein of Gastrodia elata from mainproducing areas. According to the contents of four kinds of active ingredient, the quality ofgastrodiae was evaluated in Hanzhong producing areas. It can provide the reference basesthat can comprehensive development and industrialization of gastrodia elata in hanzhong city.1. On the basis of single factor experiment, using the method of hotted water refluxextraction and applying the response surface methodology to determine the best conditionsfor extracted gastrodin and using HPLC test. Results:In the boiling water condition, purewater as solvent, the solid-liquid ratio is1:37, extraction2times and extraction time126minper times, the content of gastrodin is0.377%. Respectively comparison with the2010editionof the China pharmacopoeia of gastrodin extracted in Gastrodia elata Bl, the result shows thegastrodin content was increased by2.12times.2. The optimized processing conditions were established by using single-factor andorthogonal experimental design, which was guided by the contents of gastrodine. FreshGastrodia elata Bl. were harvested in the day, cleaned sediment, cooking25min inatmospheric pressure (105℃), cut it into (0.5-1.0cm) thick slices, dried in105℃,and thenused HPLC to measure gastrodine content. The gastrodine content was0.531%. This result is more than the traditional processing method’s, increased by43.13%.3. To determined the contents of gastrodin, gastrodigenin, polysaccharide and theprotein of Gastrodia elata from main habitats. The contents of gastrodin and gastrodigeninwere measured by2010edition of China pharmacopoeia method with high performanceliquid chromatograph. And then the contents of polysaccharide and the protein weremeasured by the sulfuric acid anthrone-ratio method and coomassie brilliant blue G-250dyeing method. The average contents of gastrodin in hanzhong′s are slightly higher than theother three regions. And the average contents of in gastrodigenin, polysaccharide and theprotein in hanzhong′s are no obvious difference from the Zhaotong, Yunnan province,Dafang, Guizhou province and Shennongjia, Hubei province. |
