Embryo implantation is a complex physiological process, regulated by many factors.Forfurther research the role of leukemia inhibitory factor and its receptor (LIF/LIFR) gene inbreeding female reproductive cycle of yak.This study proposed cloning yak LIF and LIFRgene, on which bioinformatic analysis and expression of two genes in female reproductiveorgans. CDNA based on the Indian buffalo LIFR and cattle LIF gene sequences primerswere designed, cloned LIF and LIFR gene sequences by RT-PCR methods. Bioinformaticanalysis through relevant software and online resources, combined with RT-PCR method andfluorescence quantitative RT-PCR for the purpose of genes.A complete cDNA sequence in635bp of LIF gene of yak was obtained, which contained a609bp coding region that codeda protein which consisted of202amino acids. A complete cDNA sequence in3329bp ofLIFR gene of yak was obtained, which contained a3288bp coding region that coded aprotein which consisted of1095amino acids. Cloned to LIF gene and LIFR genesequences saved to GenBank,Accession numbers are KF616828and KF616829. LIF andLIFR common expression various organ.LIFR the highest expression reproduction pregnancy cycle in Female. And the LIFRexpression was the highest of pregnancy in ovary.The gens of LIF and LIFR are conservedin evolution. Having respectively a high homology with cattle, buffalo India, The highestexpression LIFR gene in during pregnancy,might promote the embryo and fetal growth..This research continue to analyze between LIF/LIFR gene and reproductive relation laid thefoundation, And provide some theoretical basis for yak breeding. At the sametime, bioinformatics research, will be helpful for the further study of LIF/LIFR gene and itscoded protein function.
