Production And Studies On The Toxoid Vaccine And Antitoxic Serum Of The Clostridium Perfringens Type C

The toxin produced by Clostridium perfringen standard strain NCTC 3180 was inactivated by 0.3% formalin and mixed with adjuvant to prepare the Clostridium perfringen type C toxin vaccine.The antitoxin titer of colostrum and serum from vaccinated pregnant sow and its piglets was evaluated.Serum of the pig that was vaccined with the Clostridium perfringen type C toxin vaccine for many times was collected for a period of time and evaluatedThis study is divided into three parts:The first part : The preparation of the Clostridium perfringen type C toxin vaccine. The bacteria liquid was produced by recovery, enrichment and toxin production of the Clostridium perfringen. standard strain(NCTC 3180).Supernatant filtrate was made from the bacteria liquid by the filter of 0.22 microns. The toxin was inactivated by formaldehyde added in white oil and aluminium respectively to get two kinds of Clostridium perfringen type C toxin vaccine.The second part : The test of the Clostridium perfringen type C toxin vaccine. The safety,sterilization and immune efficacy of the vaccine was evaluated according to the Veterinary Biological Products Quality Standards of the People’s Republic of China.The antitoxin titer of colostrum and serum from vaccinated pregnant sow and its piglets was evaluated by indirect elisa assay and a toxin neutralization assay. The results showed that the antitoxin litter of colostrum and serum were 1:3200 and 1:1600 respectively and the neutralization titer of colostrum and serum were 1:6 and 1:6 respectively.It proved that the vaccine with no side effects was safe and reliable. It can induce satisfactory immune response in piglets.The third part : The preparation of antitoxic serum. Serum of the pig that vaccined with the Clostridium perfringen type C toxin vaccine for many times was collected and evaluated according to the Veterinary Biological Products Quality Standards of the People’s Republic of China. The indirect elia assay and a toxin neutralization assay of the mice was used toevaluate the antitoxin titer. The result showed that the serum meets the requirement of the veterinary biological products quality standards of the people’s republic of china. The antitoxin litter of the α and β toxin were 1:6400 and 1:12800 respectively. 0.1mL of the serum can neutralize 500 minimum lethal dose.
