microRNA(miRNA) is a kind of important non-coding RNA with post-transcription regulation, participating in the regulation of mammalian mammary gland development and lactation. Studies show that miRNA can complementary with 3 ’end noncoding regions(3 – untranslatedregion, 3’ UTR) of the target mRNA molecule to restrain the translation of mRNA molecule or resolve it, leading to the reduction of protein expression.It have been reported that STAT5 A, PRLR, PRL, GHR play important roles in the regulation of signaling pathways in breast milk. So this experiment is mainly to explore which miRNAs regulat the genes STAT5 A, PRLR, PRL and GHR related to cow mammary. By targetScan6.0 software to predict and analyze the potential of mi RNAs of STAT5 A, PRLR, PRL, GHR gene 3’UTR,the result respectively is miRNA-2895 and miRNA-2446(STAT5A), miR-2393(PRL), miR-135b(PRLR), miR-101(GHR).By constructing psi-check2 dual luciferase report vectors and pcDNA3.1 expression vectors of these genes, they were co-transfected into 293 T cells, respectively. After 40 hours, the cells were digested and then the dual luciferase activity was detected on a microplate reader. The result was that the ratio of firefly luciferase activity and renilla luciferase activity of psi-check2-STAT5A3’UTR, psi-check2-PRLR3’UTR, psi-check2-GHR3’UTR were improved(P<0.05), the ratio of firefly luciferase activity and renilla luciferase of psi-check2-PRL3’UTR was not significant compared with the controls. The result indicated that miR-2895 and miR-2446 may regulate STAT5 A, miRNA-135 b may regulate PRLR, miR-101 may regulate GHR. miR-2393 could’t regulate PRL.
