Wheat is one of three major important grain crops for mankind which is cultivated in most countries and regions in the world. Wheat is rich in starch, protein, is our important source of energy material and proteins. To safeguard the stability of our country of wheat production in food security and national economic development has the vital significance. Wheat acreage area is mostly distributed in the northern areas in China, such as huanghuai wheat areas, the guanzhong wheat areas, etc., these wheat areas crop failure caused by drought in sometime. Plus in recent years, the climate anomalies caused by rainfall, unequal distribution factors, making it harder to keep the wheat production in a high and stable yield. Abiotic stress, such as drought, soil degradation, salinization, has become a limiting factor of the normal growth of wheat and production in many regions. So, mining stress related genes in common wheat, developing molecular markers, and using MAS(Marker Association Selection), to cultivate good varieties, ensureing high yield and stable yield of wheat.The studies found that SKIP(Sloan-kettering retrovirus interacting protein) Protein widely involved in the process of growth and development of organisms, such as Wnt(gene name) signal pathway in animals, precursor mRNA(pre-mRNA) alternative splicing. In plants, SKIP can respond to abiotic stress. In recent years, it has been studied in Arabidopsis, rice and maize. In wheat, the gene has not been cloned. In this study, we cloned the TaSKIP in wheat, explored the expression patterns under stress conditions. The results as follows: 1、This study obtained sequence of three TaSKIP genes in common wheat Chinese Spring using homology cloning, TaSKIP-6A was located on chromosome 6A and TaSKIP-6D was located on chromosome 6D, by using the Chinese Spring nulli-tetra. 2、 Sequence analysis had shown that the coding region of TaSKIP-6A is 1824 bp, coding 608 amino acids; the coding region of TaSKIP-B and TaSKIP-6D are both 1824 bp, coding 607 amino acids. 3、Real time PCR test results show that, TaSKIP was up-regulated under drought stress condition, suggesting that the gene TaSKIP in wheat plays an important role in the abiotic stress response. 4、Construction of TaSKIP-A genetic transformation vector pBI121-TaSKIP for genetic transformation, and transformed it into Arabidopsis thaliana. We got positive plants of T0 generation, through transgenic methods to validate their adaptation or resistance to abiotic stress in the next step.
