Purpose: The study was to prepare the compound pefloxacin mesylate liquid,for the treatment of the infections caused by pefloxacin sensitive bacteria.To established for assay method by ultraviolet spectrophotometer method. At the same time, the physical and chemical properties, stability, antibacterial effect in vitro and safety and other aspects of the in-depth study, which made compound pefloxacin mesylate liquid on veterinary clinical application and provided a theoretical basis for the effective development. Pefloxacin mesylate become a broad-spectrum, efficiency, safety, reliable and new antimicrobial agents.Methods: ① According to the characteristics of liquid and component attributes, determine the liquid solubilizing agent, complexing agent and preservative in an optimum composition and proportion. ② To establish an analytical content methods, Ultraviolet spectrophotometry were measured pefloxacin mesylate and paracetamol.③Through the glare test, high temperature and low temperature test and accelerated test to investigate the stability of the compound pefloxacin mesylate liquid. ④ To test the sensitivity of compound pefloxacin mesylate liquid, Pefloxacin mesylate raw material medicine as control, compound mesylate pefloxacin oral liquid studied on Salmonella, Staphylococcus aureus, Streptococcus agalactiae and Escherichia coli antibacterial activity in vitro and determine the MIC values.⑤To evaluate the safety of compound pefloxacin mesylate liquid, the acute toxicity test were employed..Results: The main medicine pefloxacin mesylate 2%, paracetamol 0.2%, EDTA-2Na q.s.complexing agent, metabisulfite q.s. ethanol q.s.PEG400 q.s. and purified water. Ultraviolet spectrophotometry were analysis method at 277 nm and 202 nm was high precision, recovery rate and repeatability, which is an accurate, simple and reliable determination.The stability test results shows that the content of pefloxacin mesylate and Paracetamol were decreased in strong light test, although the content of them did not change in high temperature and low temperature test.The antibacterial activity of compound pefloxacin mesylate liquid was better than that pefloxacin mesylate raw medicine.LD50 of this liquid is 1453.3mg/kg for chicken. It shows that the liquid is hypotoxicity and high safety.Conclusion: The compound pefloxacin mesylate liquid which we prepared was meets the related requirements and practice,thus it should be wildely expanded in production and applied in veterinary clinic.
