The mammary gland is a complex organ with the function of lactation and the ducts and lobular are essential for the lactation, in which the ducts have the function of transporting the milk meanwhile the lobular structure participate in the synthesis and secretion of the milk. What is interesting is that the ducts and lobular are composed of mammary epithelial cells, therefore, the study of the function mechanism of mammary epithelial cells is important. Now the influence of Wnt signaling on the development of ducts and lobular has been widely studied. Many factors can effect the study of Wnt signaling on the formation of mammary gland acinus-like structure in vivo, therefore, it is important to look at the influence of Wnt signaling on mammary gland epithelial cells differentiation and function in vitro. In this study, we use the Guan Zhong dairy as the experimental material, and our aim is to have looked at the influence of Wnt activator-BIO on goat mammary gland epithelial cells biological characteristics and development. The results of the studies are listed as follows:1. The primary cells were obtained from culturing the mammary tissue and purified the epithelial cells characterized by the mRNA expression of casein and ACACA. The results demonstated that the Wnt activator-BIO promotes the formation of acinus-like structure with the mRNA expression of casein and ACACA in 2D compared,and the GSK-3 inhibitor BIO can better promote the formation of acinus-like structure and duct-like structures with the mRNA expression of casein and ACACA in 3D compared, meanwhile, mammary epithelial cells presented as the acinar structures in the presence of DMSO, but when BIO was added to the culture medium the lumen formed faster and the size of the acinar structures is bigger.2. Two types of dairy goat mammary epithelial cell with the mRNA expression of casein and ACACA were obtained by means of signal-cell cloning,including flat mammary epithelial cells(FMECs) and beechive mammary epithelial cells(BMECs). Subsequently, we studied the biological characteristics, the morphological changes of the structure and function of two types of dairy goat mammary epithelial cell in 2D compared, and compared with the BMECs, the FMECs have a higher mRNA expression of N-cadherin, meanwhile, the FMECs have the mRNA expression of ERα, ERβ and PR. However, the BMECs only have the expression of ERβ. Compared with the FMECs, the number and the size of the acinar structures which is formed by BMECs is bigger in 3D.3. The Wnt activator-BIO can promote the BMECs form acinus-like structure in 2D, however, it can not promote the BMECs form acinus-like structure in 2D. And the GSK-3 inhibitor BIO can better promote the BMECs form acinus-like structure and duct-like structures in 3D compared. Compared with the BMECs, the number and the size of the acinar structures which is formed by FMECs is smaller in the prescence of BIO in 3D.
