| micro RNAs consist of si RNAs and mi RNAs, they are a class of about 21-24 nt small non-coding RNAs, which have a great influence in Growth and development in plants and animals. micro RNAs regulate plant development by their targeted genes.mi R156 is one of the most conversed small RNAs and it is widely erpressied in plants.Through lots of researchs,mi R156 regulates plants in phase change by the targeted genes SQUAMOSA –promoter binding-like(SPL). Our laboratory has found three pre-mi R156 called Ptmi R156 a Ptmi R156 b Ptmi R156 c by transcriptome sequencing in trifoliate orange. Using the methods of STTM to build the mi R156 mimic and transformed them into trifoliate orange, for testing the mechanism of mi R156.The main results are indicated in the bellow:1. We isolated three mi R156 family genes from sweet orange trifoliate orange and precocious trifoliate Orange. According to sequence alignment among the testing species, the core sequence is highly conserved but others have little difference.Though the comparison of mi R156 in Arabidopsis thaliana Rice and sweet orange, the core sequence is truly conserved but others have lots of difference.2. Using the mi RN A Real-time PCR to test the temporal expression pattern in precocious trifoliate Orange and trifoliate Orange of mi R156. The result is that mi R156 is highly expressed in juvenile leaf and decreased when the plants got matured and it has highly difference in different organ in the flower.3. Using the methods of Short Tandem Target Mimic to build mi R156 Mimic construction and genetic transformation were carried out into trifoliate orange to explore the mechanism of mi R156. |
