Pathogenesis-related proteins such as PR1, PR2 and PR5 have been proved to be involved in the systemic acquired resistance(SAR) reaction and have been used as characteristic markers of SAR. Based on the earlier studies, in order to investigate the role of Ta Lr35PR1, Ta Lr35PR2 and Ta Lr35PR5 genes in wheat growth and Lr35-mediated adult resistance to Puccinia triticina, the expression profiles of Ta Lr35PR1, Ta Lr35PR2 and Ta Lr35PR5 genes at different growth stages of adult wheat were analyzed using real time PCR(q PCR) and western blotting(WB) respectively. Furthermore, the expression profiles of Ta Lr35PR1, Ta Lr35PR2 and Ta Lr35PR5 in tested wheat organs and the signal pathway which they might attended were identified. Main contents were as following:1. The expression profiles of Ta Lr35PR1, Ta Lr35PR2 and Ta Lr35PR5 genes induced by P. triticina at different time points were analyzed by q PCR and WB. The results showed that Ta Lr35PR1, Ta Lr35PR2 and Ta Lr35PR5 genes were induced earlier and their expression levels were higher in the incompatible interactions than that in the compatible interactions both at nucleotide and protein levels, which indicated that these three genes could be induced by P. triticina.2. The accumulations of Ta Lr35PR1, Ta Lr35PR2 and Ta Lr35PR5 transcripts increased significantly since two-leaf stage of wheat, then expressed stably at the adult stage in the incompatible interactions, and their expression levels were higher in the incompatible interactions than that in the compatible interactions and control. In addition, Ta Lr35PR1 and Ta Lr35PR2 might related to the growth of Tc Lr35 wheat while Ta Lr35PR5 changed little at the different periods of wheat development and growth.3. q PCR analysis showed that more transcripts of Ta Lr35PR1 and Ta Lr35PR2 were detected in leaves than stem and root, but the expression trends of these three genes induced by leaf rust in different organs were similar, while the expression of Ta Lr35PR1, Ta Lr35PR2 and Ta Lr35PR5 genes could be induced by chemical molecule such as salicylic acid(SA) and abscisic acid(ABA),and the expression levels of these three genes increased significantly after inoculation with P. triticina and pre-treatmented with chemical molecule. More transcripts of Ta Lr35PR5 were detected in stem than leave and root.All these results indicated that Ta Lr35PR1, Ta Lr35PR2 and Ta Lr35PR5 genes might take part in Tc Lr35 wheat adult resistance to P. triticina.
