The experiment was conducted to study the effect of adding conjugated linoleic acid(CLA) and vitamin A(VA) in layer dietary on the antioxidant index, immune parameters,fatty acids and PPAR γ and SOD gene expression in liver. In order to determine the effect of CLA and VA on the immune function and antioxidation of layers.And it might provide a theoretical basis for the health of laying hens breeding and the development of eggs rich in conjugated linoleic acid.3 × 3 double factors completely cross-group design was adopted in the experiment,in which the layers were fed the diets with different levels of Vitamin A(4000 IU/kg,8000 IU/kg,16000 IU/kg)and CLA(0%,2%,4%).Two hundred and sixteen healthful brown layers at 30 weeks old were randomly allocated to 9 treatments,each treatment had 3 replicates of 8 broilers. The trial period was 6 weeks.The results showed as follow:CLA could significantly increase total protein, globulin, albumin in serum(P<0.05),and with 4% CLA in diets,he levels of the total protein, globulin, albumin were highest among treatments. VA had significant effect on total protein,globulin in the serum(P<0.05).Total protein,globulin in the serum were increased with VA levels increasing.VA had no significant effect on albumin(P>0.05). Between VA and CLA interaction had extremely significant effect on total protein, and had no significant effect on globulin and albumin(P<0.05).CLA had extremely significant effect on immunoglobulin A(Ig A) and immunoglobulin G(Ig G) in the serum(P<0.01),and had significant effects on immunoglobulin M(Ig M)(P<0.05). The content of Ig A and Ig M in the serum were increased with CLA levels increasing,Ig G in 2% CLA treatments were higher than in 0% and 4% treatments. VA could significantly increase Ig A, Ig M and Ig G levels in serum. Between VA and CLA interaction had extremely significant effect on Ig(P<0.01).2%CLA could significantly increase the content of IL-2(P<0.01),there were differences between 0% and 4%CLA(P<0.05). VA had no significant effect on IL-2(P<0.05). Between VA and CLA interaction had no significant effect on IL-2(P>0.05). The addition 2% of CLA could significantly reduce the TNF-α in the serum(P<0.05).The TNF-α were affected significantly by the levels of VA(P<0.01),and with 4000 IU/kg VA in diets,the levels of TNF-α were lowest among treatments.Between VA and CLA interaction had extremely significant effect on TNF-α(P<0.01).The addition of 2% CLA could significantly reduce the T-AOC(P<0.05), and wouldn’t show the differences between the levels of 0% and 4%CLA(P>0.05).VA had no significant effect on T-AOC(P> 0.05), but the diet supplemented with 8000IU/Kg VA, there were an increasing trend of T-AOC.Between VA and CLA interaction had significant effect on T-AOC(P<0.05).The T-SOD were affected significantly by the levels of CLA(P<0.01),with the addition of CLA increaseing,T-SOD was increased at first and then reduced.VA had no significant effect on T-SOD(P>0.05).Between VA and CLA interaction had extremely significant effect on T-SOD(P<0.01).The GSH-PX were affected significantly by the levels of CLA(P<0.01), and with 2% CLAin diets,the levels of GSH-PX were highest among treatments. VA had no significant effect on GSH-PX(P> 0.05). Between VA and CLA interaction had extremely significant effect on GSH-PX(P<0.01).2%CLA dietary could significantly reduce MDA in the serum(P<0.05).VA had no significant effect on GSH-PX(P> 0.05),and with the addition of VA increasing,the content of MDA decreased, Between VA and CLA interaction had significant effect on MDA(P<0.05).The m RNA expression of peroxisome proliferator-activated receptor γ(PPAR γ) were affected significantly by the levels of CLA(P<0.01), and 2% CLA could significantly increase m RNA expression of PPAR γ(P<0.05).VA had no significant effect on m RNA expression of PPAR γ(P > 0.05). Between CLA and VA interaction had no significant effect on m RNA expression of PPAR γ(P<0.05).The m RNA expression of SOD were affected significantly by the levels of CLA(P<0.01). In the addition of 2%CLA,The m RNA expression of SOD were significantly higher than the 0% CLA and 4% CLA. VA had no significant effect on m RNA expression of SOD(P>0.05). Between CLA and VA interaction had no significant effect on m RNA expression of SOD(P<0.05).CLA could significantly increase contents of saturated fatty acids in liver(P <0.01). VA had no significant effect on SFA,and SFA were increased with the VA levels increasing. Between VA and CLA interaction had significant effect on SFA(P<0.01).CLA significantly decreased the content of monounsaturated fatty acids in liver(P <0.01). VA significantly decreased the content of MUFA in liver(P <0.05), with 8000IU/Kg VA in diets,the levels of MUFA were lowest among treatments. Between CLA and VA interaction had no significant effect on MUFA(P<0.05). CLA significantly decreased the contents of polyunsaturated fatty acids in liver(P<0.01). VA had no significant effect on PUFA. Between VA and CLA interaction had no significant effect on PUFA(P<0.05). CLA significantly decreased the content of unsaturated fatty acids in liver(P <0.01). VA significantly decreased the content of UFA(P <0.05), with 8000IU/Kg VA in diets,the levels of UFA were lowest among treatments. Between CLA and VA interaction had no significant effect on UFA(P<0.05).The contents of CLA isomers in liver were affected significantly by CLA(P<0.01). CLA isomers were increased with the CLA levels increasing.VA had no significant effect on CLA isomer(P>0.05),and CLA isomers were increased with the VA levels increasing. Between CLA and VA interaction had no significant effect on CLA isomers(P>0.05).The above results showed that the CLA and VA could improve immune function and antioxidation of layers,and CLA and VA had interaction effects on the immune function and antioxidant of laying hens.
