This paper take the W1-17 watermelon cultivar as material, this material was come from the Northeast Agricultural University Watermelon and muskmelon Breeding Research Laboratory, using EMS as mutagen, analyzes the germination rate, relative germination rate, relative seedling rate and growth of M0 plants after different treatment, finally determined the optimum conditions of watermelon strain W1-17 seeds. In the mutant M1 and M2 generation nurturing process, observe plant agronomic traits, to identification and analysis of the mutant phenotype.Then provide basic data for further construction of the mutant library of watermelon, broaden the watermelon resources genetic background and development watermelon functional genomics study. The main results are as follows:(1) Using six EMS mutagenesis concentration(0%, 0.2%, 0.6%, 1%, 1.4%, 1.8%), four processing time(3, 6, 9 and 12 h), analyzed the germination rate, relative germination rate, relative seedling rate after different treatment, finally determined the concentration of EMS 1%, 9 h was the optimum conditions of watermelon strain W1-17 seeds.(2) Watermelon construct optimal conditions for this mutation mutant library, this mutant library contains 120 M1 generation pedigrees and the corresponding self-M2 generation seeds. M1 generation were found: leaf black spots,white leaf spot, leaf malformation, larger leaves, leaf folds, leaf clusters, some yellow, flower malformations, abnormal petal, male no shedding, thicker stems, fruit malformation, seeds smaller, fewer seeds, seed becomes large, seedless, tendrils malformation, short stature, slow growth, etc. 20 kinds of potential variation types. The total variation frequency 18.33% up.M2 generation after selfing get about 48 different types of variation: leaf color lighter, smaller leaves, larger leaves, leaf discoloration deep within the leaf edges roll leaves no wax, the first true leaf yellowing leaves darken cleft carved leaves shallow cleft carved leaf widens, vein yellowing leaves within the volume, leaf yellowing, bisexual flowers, flowering early, immature stamens, flower malformations, deformities petals, late flowering, flowers larger, stigma variation, color lighter, smaller flowers, ovary change large, infertility, stems thicker, thinner stems, internodes shorter internodes longer, slow growth, dwarf plant height, branching mutation. Frequencies up to 20.55% of the total variance.(3) Select 10 mutation types in M2 generation plants, after the seedling cultivation,using 23 pairs of SSR primers to analyze, of which 9 copies of materials found there are variations at the DNA level.
