In order to identify genotype of known resistance loci in apple varieties, 62 apple varieties were chosed as experimental material, and 6 genotype of known scab resistance loci(Vh2、Vh8、Vb、Vbj、Vm、Vf) and 2 powdery mildew resistance loci(Pl-w、Pl-d) were detected. In order to elucidate the molecule mechanism in the disease resistance of “Qinguan” against M. coronaria, the transcriptomes of ‘Qinguan’ apple leaves were sequenced at 12, 24, 48 and 72 hours post-inoculation(hpi) with M. coronaria. The main conclusions were as follows:1. 34 varieties out of 62 cultivars in the detection of apple scab resistance loci Vh2 and Vh8 test was positive. In the detection of apple scab resistance loci Vbj, Vb, Vf, Vm, and powdery mildew resistance loci Pl-w, Pl-d, all varieties are negative. These results provided the basic principle of the parental selection of hybridization breeding for resistance.2. The comparative results of transcriptomes showed that a total of 1956 genes were differentially expressed at the 4 time points between the inoculated by M. coronaria and control samples, including 864, 828, 340 and 371 genes at 12, 24, 48 and 72 hpi, respectively, GO and KEGG analyses of DEGs suggested that the DEGS mainly related to partical cellular component,synthesis of many secondary products and varieties of defend responses were significantly enriched. Changes of cellular component contained cell wall, plasmodesma and vesicle coat protein complex-Ⅱ, up-regulation or down-regulation of secondary metabolites, including phytoalexin, anthocyanin, lignin, flavonoid, phenylpropanoid and stilbenoid might be related with resistance.
