Wheat is one of the most important food crops worldwide. The development of wheat cultivars with high yield potential and enhanced crop productivity will be necessary to meet future needs for food. The kernel size is important for grain yield and quality, grain weight is a measure of the grain size, it affects the grain yield and quality. The research of the relationship between the Ta GW2-6A and the quality has the vital significance to high yield and quality.An allelic variation with single T-base insertion at 977 bp of Ta GW2-6A was detected in Lankaodali with large kernel, designated as Ta GW2-6A-T(TT genotype), but was not found in Chinese Spring(tt genotype). According to the sequence of "T" insert,we designed primers to detect the coding regions allelic variations of Ta GW2-6A by High Resolution Melting Curve(HRM) analysis technical, and the sequence of PCR products were analyzed. To gain further insight into the role of allelic variations of Ta GW2 in wheat quality, we analyzed various quality traits and kernel traits, including the moisture, protein content, wet gluten content, hardness and kernel weight traits, on 325 recombinant inbred lines(RILs) derived from a cross between Chinese Spring and Lankaodali in Yangling and Qianxian. The results are as followed:1. Results showed that it was accurate to detect the wheat coding regions allelic variations of Ta GW2-6A by HRM analysis technical, and classified as Lankaodali genotype(TT), Chinese Spring genotype(tt) and heterozygous genotype(Tt). The PCR product sequencing results showed that the solubility curve difference is caused by the "T" base insert.2. Compared to tt genotype, the kernel length, kernel width and kernel weight of TT genotype is increased significantly. It further confirmed the allelic variation is a good allelic variation genotypes.3. The association analysis indicated that the mutated allele significantly increased the qualitytraits. The average quality traits of the kernels with genotype TT was higher than that with genotype tt.Ta GW2-6A different allelic variation can improve the quality of wheat.4. In both environments, Ta GW2-6A different allelic variation has a significant positive correlation with the quality traits except hardness. Ta GW2-6A different allelic variation can significantly increase the quality of wheat, it is possible to breed the variety of high yield and quality in breeding process.5. TaGW2-6A variation(TT genotype) was better able to improve production and quality than wild-type(tt) plants. We can guess Ta GW2 has the pleiotropic effect, or closely linked with the QTL for quality traits, it provides a molecular basis on breeding the variety of high yield and quality in wheat.
