| Foot-and-mouth disease(FMD)is a highly contagious viral disease, that may causes a great economical loss.In China, an inactivated whole-virus vaccine was used for preventing FMD compulsively.However, there are a number of limitations and concerns with its. Therefor,it is necessary to research and development alternative FMD vaccines that are secure and effective. Epitope vaccine has become one of research hot spots of FMD alternative vaccine. Cellular immunity have an important effect on immune response of FMDV infection. In addation, the main antigen sites of FMDV exist in structural protein VP1.For this reason, it is important to research CTL epitopes of FMDV that could induce cellular immunity.FMDV A/GDMM/CHA/2013 strain is a swine virus isolated from Maoming, Guangdong Province in 2013. It may have been imported from abroad. Therefore, it is critical to study cell epitopes and develop corresponding vaccines for this strain.The structural protein VP1 contains major antigenic sites of the FMDV. In this study, we get the sequence of VP1 of FMDV A/GDMM/CHA/2013 strains from Gen Bank. Then,we predict and synthesis 10 the H-2d Restricted T Cell epitopes of VP1 by the bioinformatics software. The candidate Epitopes have been trained in vitro with spleen lymphocytes from BALB/c mice that were immuned with live virus. Through lymphocyte proliferation assay,IFN-γ ELISPOT experiments and flow cytometry,we identifity that p D1(VGPTHVID)and p D4(KAPFTRLAL)are H-2Dd restricted T cell epitopes of VP1 in FMDV/A/GDNN/CHA/2013 strains. This study provides data and basis for epitopes of FMDV and the role of cellular immunity in immune response of FMDV infection. |
