| Today,cow’s aquaculture owe a large of percentage in aquaculture of our countrt.Reproduction rate is still relatively low in our country compared with the developed countries. Mainly owe to some unexplained reasons,such as abortion,stillbirth,fetal dysplasia,these restricted the numbees ofdairy cattle breeding and production performance in our country.This became one ipportant part to hinder China’s dairy. So, In order to improve our country’s dairy production,first,we need to solve the current situation of China’s dairy industry of low reproduction rate. Embryo formation plays an important role in a successful pregnancy.Studies have shown that the early placenta in the process of the formation is in low oxygen environment, with the development of the placenta, oxygen concentration has been in constant change.OPN and cyclin G1 plays an important role in the process of pregnancy.This experiment was made using different concentrations of Co Cl2 to produce anoxic environment, culture cows uterine epithelial cells in vitro, and then under the different concentration and time, using real-time fluorescent quantitative PCR detection under different conditions of OPN and the expression of cyclin G1 m RNA. Research of oxygen thereby internally cow pregnancy, to clarify the phenomenon and mechanism, and thus to improve the pregnancy rate of cows to make a certain contribution.This experiment is to use concentration and time conditions, concentration of COCl2 is respectively 400, 200 and 100 μmol/L,time for 3, 6 and 12 h.The combination of two, which is divided into nine groups.Finally through the QT- PCR determination of OPN and cyclin G1 each m RNA expression.Experimental results show that under different concentration and time(400 μmol/L, 200 μmol/L, 100 μmol/L, 12 h, 6 h, 3 h), OPN m RNA expression of measuring tool has increased, and at the same concentrations, along with the increased time table, and high concentration in the table below the expression of a lot less than the same time and low concentration; Cyclin G1 in 100 μmol/L situation in extended over time, a large increase, but in the 200 and 400 μmol/L as the table with the extension of a large drop in the world.Conclusion: under anaerobic conditions, OPN was expressed and cyclin G1, low oxygen is conducive to the expression of OPN, higher and lower oxygen concentration table a lot; and cyclin G1 under the condition of high oxygen is beneficial to express, but low oxygen conditions over time table down a lot. |
