| Ophiocordyceps formosana, an important medicinal fungus with obvious antineoplasmic activity, parasitises on the larvae of Tenebrionidae of Coleoptera. However, its genetic background of fruiting body formation and some important chemical components, remain unclear. In this paper, on the basis of analysis of mating type gene of 34 single strains from secondary ascospores of O. formosana by PCR, artificial induction cultivation process for fruiting body and composition of amino acids and nucleosides of three kinds of artificial culture were studied. The findings provide important scientific basis for future large-scale of artificial cultivation and development of O. formosana.The amplification showed that there are 14 strains with MAT1-1,4 strains with MAT1-2 and 16 strains with both mating-type genes. The result of culture experiments demonstrated that the colony diameter (7.00mm), fruiting body formation (81.25%) and the yields of strains (1.62g per canning jar) with both mating-type gene is obviously greater than that of strains with only one mating-type gene. So strains with both mating-type genes are more suitable for the study of artificial culture for fruiting body.The results of the optimal cultural conditions for fruiting bodies of O. formosana were as follows:FCC 15 selected as cultured strains, rice as the solid substrate, the nutrient solution(C),20g rice and 40mL nutrient solution per canning jar,10 mL inoculum size and temperature at 25 ℃, mycelia culture stage under the condition of complete darkness and continuous light (200-4001ux) after covering with mycelia. The fruiting bodies were harvested when they formed vigorously and almost no longer grew.The amino acid and nucleosides composition analysis of mycelia from shake culture (MSKC), mycelia from submerged culture (MSMC) and artificial culture of fruiting bodies of O. formosana (AFOF) exhibited that the content of total and total essential amino acids (33.23g/100g,10.97g/100g) in MSKC was significantly higher than that in MSMC (25.28 g/100g,8.42g/100g) and AFOF (23.87g/100,7.98g/100g). The content of adenosine (0.56 mg/g), guanosine (0.19mg/g), uridine (0.52mg/g), cytidine (0.25mg/g), hypoxanthine (0.57mg/g), cytosine (0.57mg/g) were the highest in MSKC, MSMC had the highest content of inosine (0.16mg/g) and uracil (0.21 mg/g). In addition, there were no statistically significant differences in the content of thymidine (0.04mg/g) and cordycepin (0.08mg/g) among all the samples. Compared to the relevant compositons of natural O. sinensis, some compositions of three kinds of artificial cultures of O. formosana exceeded the values of natural O. sinensis, including total and essential aminoacids, cordycepin. uracil and hypoxanthine. |
