As plant kingdom widespread class ofphenolic polymers,lignin is a very important biological macro molecules in the process terrestrial plant normal growth and development, and is closely related to human life. Cinnamyl alcohol dehydrogenase relies on the NADPH reduction cinnamic aldehyde and its derivatives, Is one of the key enzymes of catalytic lignin synthesis pathway, it works on the final step in the biosynthesis of lignin monomer. Studies have shown that, The CAD gene expression increased, rising stem lignin content, Wheat lodging enhancements. However, wheat CAD genes in vivo related research and the relationship between the lodging resistance is rare reported so far. Wheat CAD gene traits of wheat lodging is an urgent need to solve the problem.In this paper, based on cDNA sequences registered in wheat CAD GenBank (GU563724) specific primers were designed, And stem CAD activity in a group of two extreme varieties of wheat genomic DNA as template to amplify the full length, After sequencing to obtain high of consistency with the CAD gene DNA sequences from a three different wheat varieties. Combined with the species number, respectively named Y2CAD, Y38CAD, Y43CAD.The main results are as follows:1.Cloned the three gene’s DNA sequence of CAD, Respectively named Y2CAD, Y38CAD, Y43CAD, Lengths 1739bp,1767bp,1747bp.2.Y2CAD, Y38CAD and Y43CAD respectively with GU563724 sequence consistency were 35.63%,47.52% and 35.31%.3.CAD gene DNA sequence comprises at least two introns, introns one lengths of 747 bp, introns two lengths of 260 bp. Y2CAD, Y43CAD contain two introns, and Y38CAD contains only one intron.4.The one intron region appeared four SNP, at 888bp,1519bp,1528bp and 1533bp position respectively; another intron region appears a SNP, occurs at 194bp position.5.Cloning of three sequences of CAD with gramineous plants such as Bambusa multiplex、festuca arundinacea、Saccharum officinarum、Lolium、maize、Oryza sativa CAD gene sequences gathered for a class.
