| One day age Hailan-Brown Chicks were infected by Salmonella Enteritidis to investigate the effect of TLR4 signaling pathway regulated the polymeric immunoglobulin receptor. The m RNA expression of p Ig R in jejunumã€ileumã€duodenum and cecum were detected by the method of RT-PCR. The polyclonal antibody of p Ig R was prepared by the method of Molecule Cloning Technique. One group chicks were orally inoculated with 0.5mL of the bacterial suspension containing 108 CFU Salmonella enteritidis and the control group was given 0.5m L saline. Chicks from each group were randomly chosen and infected at 1, 3, 7 and 14 days. The expression of TLR4ã€My D88ã€TRAF6ã€NF- kappa B mRNA, the mRNA expression of p Ig R and protein level of p Ig R were detected by the method of real-time fluorescence quantitative PCR and western blot, respectively. The results showed as follows:(1) Successfully prepared rabbit anti chick p Ig R polyclonal antibody.(2) The jejunum tissues of chick were infected by Salmonella enteritidis, in experimental group the m RNA expression of TLR4, MyD88, TRAF6, NF- kappa B and p Ig R begun to increase at 1d, and the expression of NF- kappa B m RNA increased significantly(P<0.05); at 3d, in experimental group the relative expression of TLR4 m RNA compared with the control group upregulated(P<0.05), nevertheless the relative expression of p IgR mRNA increased significantly(P<0.01); At 14 d, the expression of TLR4, MyD88, TRAF6, NF- kappa B and p Ig R mRNA compared with the control group was not significantly difference.(3) The ileum tissues of chick were infected by Salmonella enteritidis, in experimental group the expression of TLR4, My D88, TRAF6, NF- kappa B and p Ig R mRNA begun to increase at 1d, and the expression of NF- kappa B m RNA increased significantly(P<0.05); at 3d, the relative expression of TLR4ã€My D88 and p Ig R mRNA compared with the control group upregulated(P<0.01), nevertheless the relative expression of TRAF6 mRNA increased(P<0.05); At 14 d, the expression of TLR4, My D88, TRAF6, NF- kappa B and p Ig R m RNA compared with the control group was not significantly difference.(4) Jejunum and ileum tissues in chick infected with Salmonella enteritidis, in experimental group the protein level of p Ig R compared with the control group significantly increased(P<0.01), the results show that, the jejunum and ileum tissues of chick infected with Salmonella enteritidis, the protein level of p Ig R upregulated.(5) TLR4 signaling pathway was activated by SE in the jejunum and ileum mucosa cells, and the expression of p Ig R was increased which enhanced the intestinal mucosal immune. |
