The Study On The Influence Factors Of Savoy Tissue Culture

Four different genetype savoy’7-1’’3-5’’T98-9’’W18’provided by vegetable genetics and breeding research team of Anhui Agricultural University were used as the experiment materials in this study. Investigation was carried out on the effects of the cotyledon with a short petiole adventitious shoot regeneration. The factors included the hormones ratio, seedling age, silver nitrate and different basic mediums and so on. The study about the influence of hormones ratio and density of microspore turbid liquid to the isolated microspore embryogenesis induction is also be made, and the cytological observation about different period of microspore in different length flower buds is be made.The resurts are as follows:The four materials all obtain adventitious shoots, and obtain the best adventitious bud induction conditions.The most fitting seedling age of’7-1’is 6 days, and the most fitting medium is 1/2MS+4mg/L6-BA+1.0mg/Lα-NAA+4mg/L AgNO3+0.9% agar. The most fitting seedling age of ’3-5’ is 5 days, and the most fitting medium is 1/2MS+3mg/L6-BA+1.0mg/La-NAA+6mg/L AgNO3+0.8% agar. The most fitting seedling age of’T98-9’is 5 days, and the most fitting medium is 1/2MS+4mg/L 6-BA+0.4mg/L a-NAA+5mg/L or 7 mg/L AgNO3+0.8% or 0.9% agar. The most fitting seedling age of ’W18’ is 5 days, and the most fitting medium is 1/2MS+4mg/L6-BA+0.5mg/La-NAA+4mg/L AgNO3+1.2% agar.The most fitting length of flower bud of these three different genetypes ’7-1’ and ’3-5’ and ’W18’ is 2.0-2.5mm, the most fitting length of flower bud of ’T98-9’ is 2.5-3.0mm.Smile white embro is getted from’7-1’ ’3-5’ and ’T98-9’ in isolated microspore culture.’3-5’ is getted the most smile white embro on medium 1/2NLN-13+0.3mg/L6-BA+0.1mg/La-NAA;’T98-9’ is getted smile white embro on medium 1/2NLN-13+0.3mg/L6-BA;’7-1’ is getted smile white embro on medium 1/2NLN-13+0.3mg/L6-BA+0.3mg/La-NAA. The best density of microspore turbid liquid is one bud per milliliter.
