| Guangxi common wild rice (Oryza rufipogon Griff.) contains rich genetic diversity and is a precious germplasm resource library for rice breeding. It is important to exploit and make full use of elite genes in wild rice including fertility restorer genes. In this study, ten nucleus germplasm of Guangxi common wild rice(Oryza rufipogon Griff.) which had rich of genetic resources were used as donor parents, and the cultivar 9311 was used as receptor parent. Thirteen single segment substitution lines (SSSLs) with Rf3 or Rf4 locus were developed by crossing and backcrossing. Then two double segments pyramiding lines (DSPLs) carrying both Rf3 and Rf4 loci were obtained from two crosses between two SSSLs carrying Rf3 and Rf4 locus, respectively. On this basic, the restorability of two fertility restorer genes in eight SSSLs and two DSPLs for Mengkang A (CMS-WA) and ten accessions of Guangxi common wild rice for Jinnong 1A (CMS-FA) in were were investigated. The results showed that:(1) There were significant differences in restoring abilities among these eight SSSLs:the effect of Rf3 from DP15 and Rf4 from DP15, DP30, DP50 and DP63 had stronger restorability, which the phenotyping for pollen of F1 was 86.18%-91.13%; the effect of Rf4 from DP29 had strong restorability, which the phenotyping for pollen of Fi was 81.21%; the SSSL with Rf3 from DP29 possessed weaker restoring ability; the SSSL carrying Rf3 from DP30 did not exhibited restoring ability.(2) The pyramiding effects of Rf3 and Rf4 genes varied greatly according to the donors: pyramiding line which carried strong restorability (Rf429)and weaker restorability (Rf329)from DP29 had significant additive effect for MengkangA, reached 87.50%;but the two strong restorability(Rf315 and Rf415) from DP 15 had reached high level themselves, so there was no significant additive effect after pyramiding.(3)Ten accessions of Guangxi common wild rice had different degree of restoring abilities on JinnonglA and two accessions restoring ability of GXU16 and GXU19 resched normal level, 84.30% anf 86.07% respectly, could be used as parent materials of restorer lines.The results showed that it’s effective to use the Guangxi common wild rice to obtain a number Rf3 and Rf4 alleles with strong restoring ability for the CMS-WA MengkangA. Screened two common wild rice germplasms with strong restoring ability for CMS-FA.The results of the study had enriched the rice restorer gene resources, and had important significant for further study on the wild rice restorer genes. |
