Bletilla is one of the groups of economic value of perennial herbaceous plants in orchid plants, distributing in Asia. Bletilla not only have high ornamental value, also is one of the most important Chinese traditional medicines. Especially in recent years, as people living standard rising, the influence of strong growth of the demand of the medical and horticulture market make the dosage of bletilla genus increase, export markedly increased in China. Fungi are separated from bletilla soil and bletilla root in this study, and analyzed its diversity. The research that the symbiotic fungi and bletilla seed germination experiment and bletilla aseptic germination and The effects of fungus on germination of bletilla were studied. The main results were as follows:1. The soil around bletilla gradient is dilutioned, which get 10 strains fungi. 31 strains were isolated from bletilla root. Then get 31 strains except for duplication. These strains are identified by morphology and molecular biology means. Phylogenetic tree was constructed by choosing 1000 repeats and doing the Bootstrap value analysis using Mega5.1 software according to the method of N-J clustering. The Results shows that the funguses are identified as 4 phylum 18 genus, and the diversity of bletilla fungi. They are classified into Fusarium(BJ9,BJ19,BJ50,BJ48,BJ43andBJ21), Ilyonectria(BJ27 and BJ22), trichoderma(BJ8), ChaetomiumKunze(BJ7), Exophiala(BJ25), penicillium(BJ23, BJ26 and BJ41), aspergillus(BJ39),Pyrenochaeta(BJ24), Alternaria(BJ35), Paraphoma(BJ18,BJ36), Mucor( BJ 13, BJ14, BJ15,BJ52 and BJ16), ganoderma(BJ17),Humicola(BJ47), chaeto medium(BJ49), cladosporium(BJ45), mortierella(BJ51), Tricholomaceae(BJ38),and BJ20 is identified tomentella.2. 31 strains which were isolated and bletilla seed do experiment in vitro seed germination. The results show that the existence of fungus against the germination of bletilla, and bletilla can germinate normally under natural conditions.3. According to the bletilla aseptic germination experiment, which get the following conclusions: bletilla germination under the condition of the mixture(7:3) of soil and lime is better. But the humus soil mixed simply and soil medium is very difficult to make bletilla germinate. The research makes the germination of bletilla has very realistic significance.
