| This experiment by means of somatic cell cloned embryos recipient and in vivo embryos recipient, and research on the dynamic change of reproductive hormones in different gestation period in the blood. Tried to explain reasons of abnormal pregnancy from the aspects of the level of reproductive hormone about somatic cell cloned embryos recipient, and predicted the abortion of bovine fetal, further to improve the production efficiency of the somatic cell cloned cattle. Establishing the frame of reference about the level of reproductive hormone in different gestation period, and providing reliable reference for the production of somatic cell cloned cattle. To optimize the system of the production-management of the somatic cell cloned cattle.In this study, using FSH, imported from Canada, CIDR+FSH+PGF2a method, which was a relatively mature superovulation method, was employed to obtain in vivo embryos. We established Wagyu fetal Fibroblasts through organization block adherent method, and choosed the P5 fetal Fibroblasts as a nuclear transplantation. Then somatic cell cloned reconstructed embryos were obtained through the electric fusion method.In vivo embryos and somatic cell cloned embryos transfer to the appropriate recipients. In their different gestation period, we determined the level of the reproductive hormones in the blood (FSH, LH, E2 and P) by chemiluminescence immunoassay (CLIA).We used the total dose of FSH, imported from Canada, is 380-420 IU. We obtained a total of 58 available embryos from Wagyu. We established Wagyu fetal Fibroblasts,and a total of 38 somatic cell cloned reconstructed embryos were obtained. Establishing the frame of reference about the level of reproductive hormone(FSH,LH.E2.P) in different gestation period, and made comparison with somatic cell cloned embryos recipient. In conclusion, we analysis the reasons for abortion at the level of reproductive hormone about the somatic cell cloned embryos recipient. |
