Objective to investigate the effects of Astragalus polysaccharides on bone marrowhematopoietic function in chicken and its mechanism.The text firstly researched the effectsof Astragalus polysaccharides on the erythrocyte number of peripheral blood, hemoglobincontent, blood platelet count of normal and blood deficiency Chicken in vivo experiment,in order to ensure the effect of Astragalus polysaccharides on bone marrow hematopoieticfunction. Then effects of Astragalus polysaccharides on hematopoietic growth factor(EPO,IL-1,IL-2,IL-3), hematopoietic microenvironment and proliferation and division ofbone marrow hematopoietic progenitor cells (CFU-E,BFU-E) in normal andcyclophosphamide-treated chicken were detected via combining the experiments in vivoand in vitro. Effects of Astragalus polysaccharides on bone marrow hematopoietic functionin chicken were discussed to find its way and mechanism from the cell and molecular levelto provided the scientific basis of Astragalus and its polysaccharides in the application ofpoultry production.In vivo expriment, forty chickens that prepared to feed for20days were randomlydivided into4groups of10chickens each.Group I~III were the health group withAstragalus polysaccharide, receiving Astragalus polysaccharides of100、500and1000μg/mL in water respectively. Group IV was the health group without Astragaluspolysaccharide. The14-day-old health chickens were injected with Cyclophosphamide byperitoneal cavity for6days (80mg/kg.d) to establish blood-deficiency syndrome model.Then40chickens were selected and divided into4groups of10chickens each. Group I~III were blood-deficiency group with Astragalus polysaccharide,receiving Astragaluspolysaccharides of100、500and1000μg/mL in water respectively. Group IV was theblood-deficiency group without Astragalus polysaccharide. Another10healthy chickens atage of20days were selected as healthy control group without Astragaluspolysaccharide.Groups treated with polysaccharide were administered for7days,thenblood was collected to detect erythrocyte number of peripheral blood, hemoglobin content,blood platelet count and hematopoietic growth factor (EPO,IL-1,IL-2,IL-3) content inserum. Results indicated that the erythrocyte number of peripheral blood,hemoglobincontent,blood platelet count and the content of hematopoietic factor EPO, IL-1, IL-2, IL-3in control group were higher than in model group; And the hematopoieticmicroenvironment was also improved. Experimental results indicated that Astragaluspolysaccharide can prevent and relieve the blood deficiency of chickens treated withcyclophosphamide by regulating the expression of peripheral blood, hematopoietic factorand microenvironment, which palyed function in tonifing Qi and nourishing blood. In vitro test chose the chickens from10to14days of age. Using cell culture methodto cultivate bone marrow progenitor cell of chicken, when the cell cultivate plate wascovered80%, do the research from three directions:The effects of drug serum, Astragaluspolysaccharides and he pure APS without cell factor on the growth of bone marrowprogenitor cell.According to the selected dose of drug serum and APS,the cells weredivided into5and4groups, supplied with different dose of drug serum (0.1,0.2,0.3ml) anddifferent concentration of APS (250μg/ml,375μg/ml,500μg/ml), observed everyday. Thedrug serum increased the cell proliferation ability.Middle dose of drug serum cansignificantly increase the cell proliferation ability, meanwhile, it can also prolong the celllife. But the cells supplied with the pure APS without cell factor grew slow, theproliferation ability were relatively moderated,and cannot grow normally when addeddifferent concentration of APS directly. The results indicated that the drug serum canincrease the proliferation ability of hematopoietic progenitor cell and inhibit occurence ofblood deficiency.Conclusions: Cyclophosphamide can induce the impairment of hematopoieticfunction,causing chicken blood deficiency, led to significant decrease erythrocyte numberof peripheral blood, hemoglobin content, blood platelet count and hematopoietic factorEPO, IL-1,IL-2, IL-3content; hematopoietic microenvironment are also be damaged. APScan significantly inhibit the injury of the peripheral blood, hemopoietic factor andmicroenvironment caused by Cyclophosphamide. Meanwhile APS can improve the body’sproliferation ability of hematopoietic progenitor cell from cell perspective, prevent andinhibit blood deficiency, achieve the curative effects of tonifying Qi and nourishing bloodwith Chinese herbs.
