Besides its role in regulating the formation of melanogenesis, the Agouti gene wasclosely related to energy metabolism regulation. Goat was important agricultural economicanimal. Researching and discussing the differential expression characteristics of differentviscera in different coat color goat for mRNA and its coding protein of Agouti gene not nolyprovided fundamental basis for researching the transcription regulation, upstream of gene,epigenetics, and proteomics for Agouti gene but also had great significance for moleculargenetic breeding of goat.The real-time quantitative PCR was used to investigate the expression level of AgoutimRNA in goat viscera (Heart, Liver, Spleen, Lung, Kidney) with white, brown and blackcoat color in order to explore its differential expression.2-△△C twas used to calculate Agoutigene’s Rel Quant in three levels which were in the same viscera of different coat color goatand in the different viscera for the goat having the same coat color, another level was totalrelative expression of different viscera for goat respectively. In the first level, white goat wasused as the control, the result was white goat> brown goat> black goat. There wassignificant difference of Agouti mRNA expression between white goat and black goat(P<0.01) for five viscera, and it was strongly expressed in liver of white goat. And in thesecond level, the lung was used as the control, the relative expression of mRNA was the mostin liver for white goat and brown goat, however, the most expression in black goat is kidney.And in the third level, the experiment still compared with lung, the relative transcript level ofliver was the most, and the lung was the least. It was different that the Agouti gene mRNAexpression quantity in goat viscera of different coat color.At the same time, the western blotting was used to investigate the expression level ofAgouti signaling protein(ASIP)in goat viscera (Heart, Liver, Spleen, Lung, Kidney) withwhite, brown and black coat color in order to analyse its differential expression. ASIP wereexpressed in the five internal organs of different coat color goat and a specific band (~40kDa) was detected. The expression level of ASIP was different in the same viscera ofdifferent coat color goat and in the different viscera of identical coat color goat. The basictrend was white goat> brown goat> black goat for the expression level of five internalorgans. And it kept in touch with mRNA expression in the same level. In the comparison of different coat color goat, the ASIP expression level of kidney was highest in brown goat andblack goat. For white goat, the ASIP expression level of heart was more obvious than kidney.ASIP was expressed in different amounts for goat viscera without close relationship tomRNA levels by elucidated. The result maybe related to the transcription regulation,translation regulation, protein modified and protein interaction. And the consequence alsoresulted from roles and function of different viscera, and so on.
