Sorghum is one of the world’s fifth largest food crops, with the world’s energy crisis intensifying, sorghum as a plant that have high photosynthetic efficiencyenergy began to attract people’s attention. At present, the plant breeding research has initiated the era that organic combined identification in phenotypic and molecular, through the analysis of DNA diversity in population of hybrid progeny, then study the genetic background of plants, the blindness of choice in breedingwork and the workload in breeding of hybrid group can be reduced, meanwhile,that can provid the basis for reserving least amount of material but have abundant genetic diversity as much as possible for breeding research.In this study, a set of132recombinant inbred lines (RIL) population derivedfrom a sorghum F2:3genetic mapping group between an aphid resistant cultivarnamed HN-16and a superior strain named QS through successive selfing for eight generations were used, with SSR markers,27pairs of specific primers werescreened out from173SSR primers, then we analyzed genetic variation offspringand diversity of DNA, at the same time, we can screen out lines that have similar genetic background and different in individual agronomic traits, that can provide materials for further analysis in agronomic characters. In addition, plant height of sorghum was preliminary studied,2QTLs for plant height were detected, which can be a basis for genetic improvement of sorghum with the method of molecular marker-assisted selection.1. With cluster analysis, we analyzed genetic variation offspring and diversity ofDNA that extracted from132RIL population derived between aphid resistant sorghum cultivar HN-16and aphid infect cultivar QS, the result showed that there are large genetic variations between lines of this population, we got two pairs of lines which similarity coefficient close to one and only have differenceon the aphid resistant character and plant height respectively, then we screened out a pair of primers named xtxp130with SSR labeling technique, its selection efficiency in population is86.5%. at the same time, the marker xtxp78links to plant height was found.2. By using QTL IciMapping3.2software, we statistics these screened out27pairs of primers from80polymorphism loci with corresponding format, analyze the QTL for plant height of F8population with composite interval mapping method,2QTLs for plant height were detected, they were located in chromosome6of sorghum, we named them as qPH6.1and qPH6.2respectively, the twomarkers link to plant height that screened out from RIL by the way of clusteranalysis. 3. In this study, we conducted electrophoretic analysis on13sorghum cultivars inaddition to HN-16and QS by using9markers closely chained with aphid resistance gene that screened out from location of aphid resistance gene, we canknow that sb6m2388-220, sb6m2463-527and Sb6rj2776-680markers apply toall of the cultivars, however, the applicability of sb6m4892-400marker is23.08%only.
