Porcine circovirus disease is caused by porcine circovirus type2.In this study,tissues of piglets withPMWS identified by PCR were dealt with grinding,filtrating and so on,then inoculated PCV negativePK-15cells for subcultring and identification.The results of the PCV2ORF2gene amplification, indirectimmunofluorescence assay and immunoperoxidase monolayer assay show that the virus isolated in thisstudy was circovirus type2, which were named PCV2-WL.Then we cloned and sequenced full-length geneof isolate strain in order to analyze the homology with known PCV. Comparison results show thatPCV2-WL gene contained1767bp, belonging to the European model. According to the nucleotide sequenceof PCV2and its encoded amino acid sequence analysis, it reveal that the genome sequence homology ofisolate strain and other11PCV2strains were between93.8%-99.7%, while amino acid sequence homologywere between88.5%-99.4%; Nucleotide homology of PCV1reference strains and isolate strain was only77.4%, while amino acid homology was68.3%.This study provides an experimental basis for carrying outdisease prevention and control work in Xinjiang.
