Omei treefrog (Rhacophorus omeimontis) which belongs to Rhacophorus of Rhacophoridaae distribute widely in Sichuan、Hubei、Hunan、Yunnan、Guangxi province. The complex environment among different geographical populations may block up gene flowing, causing morphology differentiation and genetic differentiation among these Omei treefrog. In order to study the differentiation and relationship of different geographical populations,our experiments focus on studying the body length and genetic differentiation.Besides, we also analyze the ancient geological events, such as Quaternary ice age, which influences on the differentiation of different geographical populations.1、The result of body length:We measured and compared body length of473specimens (367males and107females) from8populations. We utilize Mann-Whitney U test to analyze sexual dimorphism of the geographical populations from Sangzhi county Hunan province and Baoxing county Sichuan province. For body length from different geographical populations, we use clustering analysis in SPSS19.0. The result of body length:(1) There is significant difference between male and female in body length of the two geographical populations (P<0.01).(2)The result of clustering analysis is Hunan、 Guangxi and Yunnan geographical populations males length are lager than others (included Hubei and Sichuan geographical populations). They are cluster respectively.2、The result of genetic differentiation:We use combined sequences of Cyt b and ND2gene to study the genetic relationship between different geographical populations.There are204genetic samples from8geographical populations and we obtain4results:(1)8populations of Omei treefrog have high genetic diversity, existing119variation locus and45haplotypes, including3shaded haplotypes (between LGL and ZPP).(2) There are distinct genetic variations among different geographical populations through genetic differentiation coefficient. Except Fst between ZPP and LGL, Fst among geographical populations are from0.5to0.99(more than0.25).The result of gene flow also demonstrates genetic variations among different populations are obvious.(3)The result of phylogenetic tree analysis (MP method and Bayes method) can be arranged into two highly supported groups:branch A and B. Two phylogenetic groups are formed respectively. Branch A contains Sichuan populations (include4geographical populations) and Hubei population. Branch B contains Hunan population、Guangxi population and Yunnan population.(4)According to molecular clock, branch A and branch B are estimated to diverge at2.6-3.15Ma BP, which may be caused by Quaternary ice age.(5) A unimodal mismatch distribution of clade B1(Hunan and Guangxi populations)has been observed, which suggest the population has expansion, while other populations don’t have.
