Red rice is selected as the research object, germination parameters of the red rice is optimized, the red rice is biforticated with the exogenous zinc. With different concentrations of zinc sulfate solution to soak the red rice, the effect of the concentration of zinc sulfate solution on the germination rate of the red rice is studied. Treating the germinated red rice which was soaked by the zinc, nutrient substance were separately determined. We studied the influence factors during the germination process and different concentrations of zinc sulfate solution on germination of nutrients in the red rice. The zinc content in the soaked red rice was determined by FAAS method. The nutritional value in the zinc-rich red rice was preliminary assessed according to the DRIs(Chinese Dietary Guide,2000).(1) By orthogonal experiment, germination conditions of the red rice is optimized, it is concluded that the best condition is (soaking time for12h, soaking temperature33℃, germination time for24h, germination time35℃),the influencing factors on the germination rate was as follows: soaking time>soaking temperature>germination temperature> germination time. Using zinc sulfate solution to soak the red rice with optimized parameters, the effect of different concentrations of zinc sulfate solution on red rice germination rate was studied. The results showed that25mg/L zinc sulfate solution could improve the germination rate of the red rice, high concentration of zinc sulfate solution(>150mg/L) will reduce the germination rate.(0-100mg/L) zinc sulfate would be the suitable solution for red rice sprouting.(2) Nutrient substance and mineral substance content were separately determined in the red rice when the germinated red rice which was soaked by the zinc sulfate solution. The results showed that the germination process can significantly improve the nutrient content of the red rice. Compared with the red rice which was not germinated, in the germinated one which was soaked by different concentrations of zinc sulfate solution, the content of GAB A, vitamin C, protein, and reducing sugar increased to133%291%,131%~176%,9%~28%,221%~262%respectively; When Zinc sulfate solution’s concentration was higher than150mg/L, GABA content decreased significantly, there are significant differences compared with the control group(p <0.05), high concentration of zinc exerts a depressant action on the production of GABA; Along with the change of zinc concentration both the protein and vitamin C content was first increased and then decreased,while the content in general was higher than that in the control sample; The content of reducing sugar exists significant differences in individual zinc concentration, such as25mg/l,50mg/l,100mg/l,250mg/l.(3)The content of mineral elements in red rice exists significant differences, with decreasing order as follows:Mg> Mn> Fe> Cu. No matter the red rice germinated or not,the mineral content had no obvious change. Compared with the control sample, the content of iron was significantly influenced after soaking the red rice in the zinc sulfate solution. Biofortification with Zn sulfate solution could increase the content of iron in the germinated red rice, which could generally increase to19.2%~45.4%. The content of Cu, Mg, Zn existed significant differences when soaking the red rice in the individual concentrations of Zinc sulfate solution. The Cu content significantly reduced when Zn concentration of culture solution was25mg/L.25mg/L,50mg/L,100mg/L zinc sulfate solution could significantly increase the content of Mn.200mg/L zinc sulfate solution can significantly reduce the content of Mg.(4)The content of zinc in the germinated red rice was determined by FAAS method.The result showed that the content of zinc was significantly increased after soaking the germinated red rice in the zinc sulfate solution, when Zn concentration of culture solution ranged from25mg/L to250mg/L,the zinc content in the germinated red rice ranged from116.3to337.9mg/kg, compared with18.5mg/kg in the control sample,it was increased to528%~1726%.The correlation between the concentration of exogenous zinc and the accumulation of zinc in the red rice was (r=0.972), which meaned that they were highly relevant. According to the Chinese residents’dietary mineral zinc Recommended intake, we calculated the demand for zinc of adult, who was taking the zinc-rich germinated red rice powder maked by25~100mg/L Zn culture solution. In order to meet the daily needs, the intake of Men should reach(50.6~107.4g) and women should reach(30.4~64.5g).
