In China, beef is an indispensable kind of meat varieties of daily diet. It’spopular for the public becaue of the high nutritional value. Nonetheless the output ofbeef is still reducing in China in recent years. The safe and high quality beef is moreand more needed. So how to increase the output of beef is a very important question.As the main cattle breeds of meat and dairy, Chinese Simmentals has been widelyused in the production of the cattle industry in our country. It also has a goodproduction performance and growth performance. Following the rapid development ofmodern biotechnology, it has become the hot research area means of beef cattlebreeding to adopt the molecular breeding technology for the genetic characteristics ofmeat traits and even the improvement and the expansion of group selection. In thisstudy, regarding DHCR24(3β-dehydro-cholesterol-24reductase)expressed by bovinemeat quality and carcass traits gene as the object of study, we establish the genepolymorphism of Chinese Simmental DHCR24gene based on the PCR-RFLP reactioncondition optimization, and then proceed further study for the differences ofcorrelative indexes for the DHCR24gene single nucleotide mutation of Differentgenotypes between individuals from meat quality and carcass traits in bovine. Specificresearch contents and results are as follows.This experiment is carried out with50Chinese Simmentals as test material bygetting the DHCR24different gene types by PCR-RFLP. On the base of optimizingthe PCR reaction conditions, reaction system, reagents and enzyme digestion system,conditions and reagent, we establish the gene polymorphism of DHCR24gene basedon the PCR-RFLP reaction condition optimization.445Chinese Simmentals weregenotype detection by using that method for analysesing the correlation between meatquality and carcass traits. The results showed that there were significant differencesbetween the different genotypes of the DHCR24gene and the variety of meat qualityand carcass traits. In445Chinese Simmental populations, allele G was the dominantallelic gene and GA was the dominant genotype. Mutant type homozygous AA genotype in carcase depth and carcass traits such as long and fat coverage has obviousadvantages; Heterozygote GA in hind leg, fat colors and marbling has obviousadvantages; Wild type homozygous GG in backfat thickness, leg thickness,circumference of hind leg, eye muscle area and waist thickness, has obviousadvantages.By this research we can diagnose the change of bovine DHCR24gene Singlenucleotide polymorphism and meat traits, to povide specific operational approach andreference frame of Chinese Simmental early gene diagnosis, choices, MAS of cattlesand molecular breeding.
