Endosymbiontic microbes are widely distributed in nature, exist in specific parts of their hosts, especially Wolbachia. Endosymbiontic affect their hosts in a variety of ways in order to favor their own transmission, and have evolved over time as a result of interactions between the host and the bacterium. Because endosymbionts can not be artificially cultured, in current, most of study on endosymbionts focused on infected detection and phylogenetic analysis. There have a conceive that we can control the host’development and reproduction by symbiotic bacteria, to achieve biocontrol. The last20years have witnessed an explosion of interest in reproductive parasites of endosymbionts.By using specific primer gene of Wolbachia、Cardinium and Spiroplasma, we measured the infection status of Wolbachia in Chilo suppressalis (Walker)(Lepidoptera:Crambidae), from14locations(Xinyang, Xishui, Tongcheng, Chibi, Xiaogan, Qianjiang, Jiangling, Jingzhou, Dangyang, Yicheng, Laifeng). The results showed that8geographical populations were infected with Wolbachia, the highest rate was100.0%in Wuhan Xinzhou. There have5geographical populations were infected with Cardinium, the highest rate was35.7%in Yicheng. There have7geographical populations were infected with Spiroplasma, the highest rate was50.0%in Xishui. The infection rate of Wolbachia (Xinyang, Xishui, Qianjiang, Yicheng) in different years were compared, we found there were no difference or no significant difference between2011s and2012s. Then the description of the infection rate of Wolbachia is relatively stable in natural populations.Chilo gene vivo infection were compared samples of the three species bacterial gene sequences exhibit a high consistency with100%similarity. We build the Maximum Parsimony Tree by the sequences of three endosymbionts in this study and other sequences downloaded from Genbank for phylogenetic analysis.Then, aligned the obtained sequences with the published ones, the phylogenetic relationships among the wsp and16S rDNA gene found in Chilo suppressalis were established respectively. The MP tree shows that, Pieris rapae、Coliaserate poliographus、 Tetranychus urticae fall into a same clade. Cardinium in this study and which in Oppiella nova、Spiroplasma of C.suppressalis we detected with the symbionts reported from Fannia manicata closely related with each other.
