Gold immunochromatography assay technology is a rapid solid membrane immuneanalysis using nitrocellulose filter membrane as carrier, and developed on the basis of goldlabeling technique, immunological diagnosis techniques and chromatography analysistechniques. GICA had been the one of immunological detection technology which weresimple, rapid, sensitive and specific, which has great developmental potential and wideapplication prospect. It has been the focus of research and application in Multi-Domain.1. Development of a rapid detection of Duck Tembusu Virus using colloid goldimmuno-chromatography assaySince April2010, a novel contagious disease in ducks and geese, with egg drop, feeduptake decline and neurological signs, caused by a newly emerged virus has spread aroundEastern China. A rapid method for detection of duck Tembusu virus diseasewas developed.The method represents a immunochromatographic assay. The contact between the sampleand the test strip with immobilized immunoreagents initiates the fluid movement along themembrane components of the test strip, immunochemical reactions, and the formation ofcolored bands. The test indicated that: no cross-reactivity with AIV and ND. The preparedcolloid gold immuno-chromatography test strips are simple, rapid for clinical applicationafter further study.2. Development of a rapid and quantitative detection of Foot-and-Mouth disease virus Asia1type using colloid gold immuno-chromatography assayIt is of great significance to diagnose the foot-and-mouth disease early, which is ahighly contagious disease. Molecular biological detection methods with many advantagesand disadvatages, such as the Real-time fluorescent PCR, the multiple PCR, the nucleicacid hybridization, the gene chip, the loop-mediated isothermal amplification and thenucleic acid sequence-based amplification, have been widely used in the diagnosis andclassification. The analysis and comparison adout the detection afficiency, sensitivity,advantages and disadvatages as well as the research status at home and abroad werereviewed in this study and provided the reference for the moleculer biology detection offoot and mouth disease virus applied to in-depth development.To develop a rapid and quantitative colloid gold immuno-chromatography assay ofAsia1type Foot-and-Mouth disease (FMD) virus. Aiming at detecting FMD virus of Asia1type quantitatively, the colloid gold-based immuno-chromatographic test strip wasprepared and the standard curve was constructed. Testing the laboratory samples andfield samples, results showed that this assay had good sensitivity and specificity. Thismethod could accomplish qualitative and semi-quantitative detection in15minutes; thesensitivity was up to0.78μg/mL with a linear ranging from0.78μg/mL to7.84μg/mL. The coefficient of variation was0.2%to5%, and the recovery reported to be91%to102%.Good reproducibility and accuracy of this method was demonstrated. The colloidal goldimmuno-chromatographic assay is rapid, specific, sensitive, accurate, worthy torecommand its clinical use.
