Porcine kobuvirus was firstly amplified by Pankovics P in Hungary from the piglets association with diarrhea. The porcine kobuvirus were detected in China, Korea, Thiland, Japan and US, more and more area were reported to amplify the positive porcine kobuvirus.In order to know the VP gene of the porcine kobuvirus isolated in Fujian province, the VP gene of porcine kobuvirus was amplified by RT-PCR method from tissues of piglets association with diarrhea from Fujian and then sequenced.The results showed that the porcine kobuvirus VP gene was2529bp in length, coding an open reading frame (ORF) with843amino acids, which combined with three structure protein:VP0, VP3and VP1.The nucleotide sequence and the amino acid sequence deduced from the gene were analyzed by the bioinformatics software. Compared with the PKV VPO genes published previously download from GenBank. The results demonstrated that the porcine kobuvirus structural protein VPO was1098bp in length, coding an open reading frame (ORF) with366amino acids. The molecular weight, theoretical isoelectric point, the instability index (Ⅱ), Aliphatic index and the Grand average of hydropathicity (GRAVY) of porcine kubovirus nonstructural protein VP0were38.489kD,6.71,34.65,71.72and-0.213, respectively.The nucleotide sequence and the amino acid sequence deduced from the gene were analyzed by the bioinformatics software. Compared with the PKV VP3genes published previously download from GenBank. The results demonstrated that the porcine kobuvirus nonstructural protein VP3was669bp in length, coding an open reading frame (ORF) with233amino acids. The molecular weight, theoretical isoelectric point, the instability index (Ⅱ), Aliphatic index and the Grand average of hydropathicity (GRAVY) of porcine kubovirus nonstructural protein VP3were24.144 kD,5.56,32.50,85.38and0.131, respectively.The nucleotide sequence and the amino acid sequence deduced from the gene were analyzed by the bioinformatics software. Compared with the PKV VP1genes published previously download from GenBank. The results demonstrated that the porcine kobuvirus nonstructural protein VP1was762bp in length, coding an open reading frame (ORF) with254amino acids. The molecular weight, theoretical isoelectric point, the instability index (II), Aliphatic index and the Grand average of hydropathicity (GRAVY) of porcine kubovirus nonstructural protein VP1were27.006kD,4.35,46.6,66.14and-0.055, respectively.The nucleotide sequence and the amino acid sequence deduced from the gene were analyzed by the bioinformatics software. Compared with the PKV VP genes published previously download from GenBank. The sequenced gene shared the highest homogeneity with XX strain at86.7%, and the lowest homogeneity with swine/S-1-HUN/2007/Hungary strain at83.7%. The porcine kobuvirus shared two different branches in the phylogenetic tree, and the China isolated at the two subbranches.
