Avian influenza is one of the main diseases threatening the poultry industry in China,it has many subtypes, The study is aim to establish operating system of H9N2subtypeavian influenza virus in reverse genetics and to rescue avian influenza recombine activevirus of H9subtype which is suitable for the production of vaccine. A kind of virus whichhave a common way of gene combination with H9N2subtype virus by operative techniqueof reverse genetics including6gene segments of H1N1subtype(PB2、PB1、PA、M、NPand NS) and2gene segments of H9N2subtype(NA and HA) have constructed, as donorsof gene segments. We investigate into growth characteristic of the recombination virus in293T and MDCK cells and evaluate security and stability of the recombination viruspreliminarily. We have recured the recombination virus of H9N2subtype which isconsidered as the vaccine candidate strain. The results of study indicate that therecombination virus is provided with stability of heredity favorably. The successfulestablishment ofH9N2subtype AIV reverse genetic operating system laid a foundation forthe study on pathogenic mechanism, mechanism of the spread, and virus gene function ofavian flu virus. The save on recombinant strains of AIV opened up new avenues fordeveloping H9subtype avian influenza vaccine.
