In vertebrate,endodernal organs pancreas and liver play a key role of digesting,and they coordinately control body metabolism.During liver and pancreas development, cell differentiation,organ morphogenesis and other events are important and complicated,If one of these development events is abnormality,it will result in malformed organogenesis and serious diseases.In our country, the rate of liver diseases such as hepatitis, cirrhosis, hepatocellular carcinoma and pancreas diseases including diabetes are high,making great threaten to human health. So thes years, molecular mechanisms underlying liver and pancreas development have been intensively studied. A series of transcription factors and signal pathways involved in fate determination and cell differentiation have been identified. However, how these transcription factors and signal pathways regulate fate determination and cell differentiation remains a major question.In our lab, the zebrafish were mutagenized with ENU to induce mutations in the spermatogonial cells, afer screening,we identified a mutuant named sbd with defective in liver and pancreas development.In our study,we research the phenotype of mutant sbd futherly,and the mutant displayed three categories defects:(-)At the early stage,liver specification is perturbed;(二) Exocrine pancreas are almost defect;(三) endocrine pancreas is displaced. In zebrafish,we usually use positional cloning to clone the mutated gene.The SSLP markers for rough mapping in initial mapping panel must have polymorp hsim.In our study, we checked712SSLP markers and screened222useful on e to make the initial mapping panel. Since the genome size of zebrafish is appro ximately2500cM, the average marker spacing is about11.2cM in the initial ma pping panel.There is a disadvantge that the limit for detection linkages is appro ximately36cM, but there is a gap about47.5cM,and greater than36cM in the initial mapping panel.My study lay the foundation of mapping mutation genes i n our lab tomorrow..
