| In this chesis,3experriments were carried out to evaluate the feasibility of a self-established semi-in vivo method for mycotoxin absorbents text.The supernent of stomach and intestine digesta from heathy adult sow were used as liquid medium in new method of distilled water in the traditional absorbency test.The absorption rates of AFB1and ZEN of11laboratory prepared mycotoxin absorbents were compared between new method and previous isthermal method.The effect of chemical modification of betonite and zeolite with new material on the absoption rate of AFB1and ZEN was evaluated using the mentioned method.Exp.1:The optimization of HPLC parameters for AFB1and ZEN analysis were optimized. Select the parameters of extraction, purification and elution conditions, and under this condition, AFB1and ZEN show good linearity in the range of lng/ml~10vg/mlã€20ng/ml~50vg/ml respectively and the detection limits were0.16ng/ml and2ng/ml, correlation coefficients were1and0.999. Relative standard deviation (RSD) of Day precision density for AFB1:2.37%; ZEN:4.50%, relative standard deviation (RSD) of inter-day precision density for AFBl:2.74%; ZEN:8.84%. The rate of recoveries for AFB1is in the range of89.10~110.3and ZEN is98.7~104.1%.Exp.2:According to the quantity of ion exchange, select one kind bentonite and one kind zeolite meeting our demands from a variety of non-metallic mineral, use two kinds of organic modifiers to modify them respectively, then the absorbents we got were P1ã€P2ã€P3ã€M1ã€M2ã€M3ã€N1ã€N2〠N3, also evaluate its adsorption rate to AFB1and ZEN in vitro The results showed that:when the densities of AFB1and adsorbent were10μg/ml and lmg/ml respectively, the adsorption rates of9adsorbents were as follows:64.45%,70.77%,92.93%,76.70%,75.08%,87.14%,63.70%,61.00%,56.73%; And when the densities of ZEN and adsorbent were10μg/ml and lmg/ml respectively, the adsorption rates of9adsorbents were:56.67%ã€86.30%ã€97.41%ã€12.77%ã€31.03%ã€78.36%ã€15.33%,10.18%ã€51.18%.Exp.3:Evaluate the adsorbent effects of P3, M3, Y1, Y2to AFB1and ZEN by simulating the circumstance of gastrointestinal tract semi-in vitro.The results showed that the adsorption rates of4adsorbents in the gastric environment on AFB1were as follows:72.10%,84.18%,83.17%and66.01%; in the small intestinal fluid the absorption rates are as follows:78.15%,88.08%,88.12%and67.34%. Four kinds of adsorbents in the small intestinal fluid environment of the AFB1adsorption rate were significantly higher than in the gastric fluid environment. In the gastric juice environment adsorption rates to ZEN were as follows:85.35%,18.41%,16.20%and12.98%; in the small intestinal fluid the absorption rates are as follows:39.75%,25.15%,21.40%and24.85%. Adsorption rate of four kinds of adsorbents to ZEN in the small intestinal fluid environment was significantly higher than gastric fluid environment. Between the two kinds of environments, the adsorption efficiency of adsorbent P3to ZEN are the best, but in the small intestine the absorption rate was only39.75%.According to the investigation findings of the contamination on grain and feed at home and abroad, design the actual toxin concentration and under the conditions of adsorbent dosage, the adsorption rate of adsorbent to ZEN and AFB1, study the effect of interaction between the two toxins on the adsorption efficiency. The results showed that the adsorption efficiency of a toxin reduced as the increase of the concentration of the other toxin, but the difference was not significant. |
