Hotan is a city located in the southernmost part of Xinjiang province, whose specialgeographical location gives birth to the special ecological system. Spontaneously there is a famousprimitive poplar desert forest ecosystem growing in the environment, which is under thoseconditions: scarcity, high temperature, drought and high radiation. There contain special microbialcommunities in the soil. Thus, this article does some researches on Actinomycetes resources fromits original special environment as experimental materials of the soil of Populus euphratica forestin Hotan area of Xinjiang. Its purpose is to find more original and unique actinomycetes speciesresources; while looking for a more valuable actinomycetes species is helpful to explore the typesand functions of their metabolites.Isolation of Soil Actinomycetes and Screening of Antagonistic StrainsSoil samples were collected from mixed samples (11collection points) of Populus euphratica inHotan area of Xinjiang. Using10kinds of isolation medium was to separate3genera including38species of actinomycetes. These three were Saccharothrix, Nocardia and Streptomyces, and themost important of species is Streptomyces containing35types. The Gram positive(Staphylococcusaureus, Staphylococcus epidermidis), gram negative (Escherichia coli) pathogens, humanpathogenic fungi (Candida albicans), agricultural pathogenic fungi(Fusarium oxysporum f. sp.Vasinfectum, Verticillium dahliae, Alternaria alternate Keissle)from the isolation andidentification of38species of actinomycetes were selected as target bacteria to screen bacteriawith antibacterial activity. Finally, it is found that18species of actinomycetes have differentantimicrobial effects on the target pathogens, including13species having inhibitory effects on S.aureus,9types having inhibitory effects on S. epidermidis, as well as4having inhibitory effectson C. albicans and2species of that having inhibitory effects on Alternaria alternate Keissle;while none of the strains had inhibitory effects on E. coli and V. dahliae.Potential new species classification of multiphase TRM43006Using methods of multiphase classification, it can identify Populus euphratica actinomycetesTRM43006from the soil of Xinjiang Hotan area of Populus euphratica forest systemically andspecifically, according to the characteristics of microbial morphology, genotypes, chemical andphysiological and biochemical characteristics of the whole-cell characteristics, and determine itstaxonomic status.TRM43006as a new species of the genus Streptomyces, named Streptomycesindologenes.Research on the metabolites of TRM43006An isolated Actinomycetes Strain TRM43006from Hotan area of Populus euphratica forestsoil of good antibacterial activity was studied on its metabolites. This experiment was applyingmacroporous resin accumulation, thin layer chromatography, silica and sephadex LH-20gel column chromatography method to separation and purification, three compounds were isolatedand determined as1H-indole-3-propanal-β-oxo (1), Bis(2-ethylhexyl) benzene-1,2-dicarboxylate(2),13-methyltetradecanoic acid (3). These compounds were isolated from Streptomyces for thefirst time and compound3significantly inhibited biofilm formation of Staphylococcusepidermidis.
