In this study, Ctenopharyngodon idellus、Squaliobarbus curriculus and their hybrids F1as experimental material, the use of conventional karyotyping methods are described in its karyotype features and compare and study the results of comparative analysis by karyotype, cytogenetic analysis from the perspective of the initial hybrid offspring fertility and detect whether the hybrid offspring of chromosome mutation, and thus enrich the contents of the cell genetics, cytogenetics and laid the theoretical foundation for the cultivation of new varieties resistant grass. The results of this study were as follows:1. intraperitoneal injection with PHA and short-term kidney tissues cultivation method used for Ctenopharyngodon idellus、Squaliobarbus curriculus and their hybrids F1chromosome preparations, and achieved good results.The results showed that the diploid chromosome numbers of these four fishes were all48, the larger chromosomes of which were all in the group of submetacentric chromosome. the karyotype of Ctenopharyngodon idellus was:2n=20m+24sm+4st, NF=92;the karyotype of Squaliobarbus curriculus was:2n=14m+30sm+4st, NF=92;the karyotype of the direct cross F1was:2n=18m+26sm+4st, NF=92;the karyotype of the reverse cross F1was:2n=26m+22sm, NF=96.While the secondary constriction, heterosome and satellite chromosome were not observed by routine giemsa’s staining method.2. By comparison analysis found that the direct cross F1karyotypes between their parents, and Ctenopharyngodon idellus、Squaliobarbus curriculus karyotype high similarity and the direct cross F1karyotypes more biased in favor of female carp, showing a partial maternal inheritance characteristics;the reverse cross F1karyotype is not within the parental karyotypes range, and the lack of a proximal portion of centromeric chromosome parents have, probably due to the distant hybridization is a genetic relationship between different genomes combined, so the hybrid appears abnormal mitotic phenomena, leading to chromosomal aberrations.3. The experimental results showed that Ctenopharyngodon idellus、Squaliobarbus curriculus and their hybrids F1chromosome number were48, in line with the number of chromosomes dace fish subfamily distribution, comparing the results with inter carps subfamily showed that Ctenopharyngodon idellus、Squaliobarbus curriculus and the direct cross F1and carps in bream subfamily、xenocyprinae subfamily and silver carp fish karyotype were high similarity, indicating with a closer relationship between them may belong to the same clade.4.By comparison analysis found that Ctenopharyngodon idellus、Squaliobarbus curriculus and the direct cross F1karyotype similarity was higher、from the perspective of a preliminary forecast cytogenetic the direct cross F1with fertility.
