| In this study, through the use of sand culture and watered nutrient fluid and field planting, took flue-cured tobacco variety K326and Yun87as material. Mainly studied after multi-topping, using different concentration of glucose oxidase in different time on flue-cured tobacco growth, alkaloid accumulation and the influence of the tobacco leaf quality, the result is as follows:1. Studying the effect that the key enzyme activity of flue-cured tobacco carbon and nitrogen metabolism and the nicotine content in different ways of injure through pot experiment Results show that the injure situation of tobacco plant, nicotine in tobacco leaves began to accumulate, T1and T4treatment, respectively18.125times and20times the level of CK. Compare with T2, treatment the nicotine of T3treatment is lower and it is87.30%on T2treatment; Nitrate reductase activity in multi-topping and pest injure cases, the decline in time forward. After120h, the Nitrate reductase activity of T3treatment descend obviously and it is29.03%comparing with CK treatment; Amylase and invertase activities began to decline after injury, pest injure treatment decline rate is lower than other ways of injure. Overall, the injure mechanism is the important factors that affect the synthesis of tobacco nicotine, comparing various injure mechanisms, pest injure mechanism is slightly different with other common injure mechanism.2. The different consistence of glucose oxidase (GOD) solution for flue-cured tobacco K326after topping and scrawling. Detecting the treated tobacco and the treated tobacco’s some chemical elements and the nicotine in leaves and roots. Analyzing and discussing effects of the different consistence of GOD for the nicotine and other chemical elements. The result shows that the different concentrations of GOD inhibit the accumulation of the nicotine in roots and leaves after topping in different degree. After120h, the2mg/ml GOD has the most outstanding result for leaves, and it is75.89%comparing with the nicotine of the untreated tobacco. The lmg/ml GOD has the best result for reducing the nicotine in roots of tobacco, and it reduce28.67%. In the same time, every concentrations show their inhibitions for the total nitrogen, but they have no obviously difference. And the GOD has no obviously effects for the growing and other chemical elements in specific consistence.3. In order to study the glucose oxidase (GOD) solution for accumulation of flue-cured tobacco alkaloids inhibition effect and quality of tobacco leaves, and daubing lmg/ml and2mg/ml GOD solution respectively after topping in0h,2h and4h. Analyisising the effects of alkaloids and qualities are caused by the different concentration GOD and time of using. Results show that injecting GOD solution to the upper leaves and middle leaves of Yun87flue-cured tobacco have different degree effects of inhibition for the accumulation of the alkaloids. The nicotine that upper leaves of T1treatment has descend obviously, it is80.46%of CK treatment. In Reduced nicotine, the upper leaves of T1has the best effect on inhibition, comparing with CK treatment, it descend23.08%. In neonicotine, the upper leaves of T2has the best effect on inhibition, it is65.82%of CK treatment. In Myosmine, the upper leaves of T6has the best effect on inhibition, comparing with CK treatment, it descend36.55%. In addition, it has no adverse effects that injecting GOD solution, and it the most of treatments have effects that controlling nitrogen content and increasing potassium content. The dates of agronomic traits and economic traits also show that injecting GOD solution has no adverse effects about them and some treatments are better than CK treatment on them. |
