Streptococcus suis is gram positive bacteria, it is an important pathogen of the disease that can cause animals disease, it can cause meningitis, myocarditis, sepsis and other symptoms. The Streptococcus suis disease are widely distributed, with the de velopment of the factory in pig, streptococcus suis disease has become one of the important diseases in pig industry. This disease can happen all the year round, the disease incidence and mortality rate is higher, bring huge economic losses to famers. Not only that, it can also cause human infection, with the disease, it can even cause human deaths, seriously threatening human public health security. For the Streptococcus suis disease prevention and control is still mainly depends on the application of antibiotics, but lots of abuse of antibiotics for a long time will lead to drug-resistant strains appear constantly, make disease outbreaks. Vaccine plays an important role in disease prevention, select vaccine is safe, reliable, able to make the animal body produces antibody levels to high resistance to bacterial infection, so as to achieve prevention purposes. But recent studies of group D streptococcus suis vaccine is less. The purpose of this experiment is to apply the orthogonal design, to optimize group D Streptococcus suis vaccine immune condition, designed to provide experimental basis for streptococcus suis active immune prevention.This experiment by selecting the antigen dose, immune pathways and time interval of three factors, each factor of three levels, according to the L9(34) orthogonal table of immune animals, measured under different combination of immune, rabbit serum agglutination titer, IgG levels, the spleen weight coefficient of rabbit, cloning of specific B cell count and percentage of B cell proliferation, so as to select the best combination of immune. By analyzing the experimental results, considering the experiment to discuss the results of application in the real environment, and use cost, immune problems, we finally choose the best immune condition combination is : the antigen dose select 1 x 108 cfu/mL, the immune way choose subcutaneous injection, immune time interval choice for two weeks.
