Objective:Taking tea saponin as the candidate drug, through the research on the influence of tea saponin to Marc-145 receptor CD163 and Vimentin gene synthesis and protein expression, as well as tea saponin whether can affect PRRSV infection by cell endogenous apoptotic pathway, to explore the antiviral mechanism of tea saponin.Methods:Through the qRT-PCR technology to research the influence of TS to cell receptor CD 163 and Vimentin gene synthesis level, At the same time,using Western blot technology to detect the influence of TS to cell receptors CD 163 and Vimentin protein expression. In addition, the research on TS to the activation effect of Caspase-9 which is cell endogenous apoptotic pathway promoter by Western blot technology make a preliminary explore antiviral mechanism of TS.Results:1.qRT-PCR results show that on the PRRSV infected Marc-145 cell TS can significantly inhibit the gene synthesis of cell receptors CD 163 and Vimentin.2.Western blot results showed that TS can significantly inhibit the expression of cell receptors CD 163 and Vimentin protein.3.Western blot results showed that TS induced activation of Caspase-9 which is cell endogenous apoptotic pathway promoter.Conclusion:1.On Marc-145 TS can significantly inhibit PRRSV enter cell to anti-PRRSV.2.TS may through cell endogenous apoptotic pathway and in promoting the early apoptosis way achieve anti-PRRSV effect...
