Flowering is a very important agronomic trait as a symbol from vegetative growth to reproducative growth plants. Our team found a dwarf and early flowering mutant line from the Datong Honggu, named mopl.Then we investigated its main agronomic traits such as the height of strain,the length of sprinkle,the number of nodes on main stem against with normal Datong Honggu.The tested result of mutants of main agronomic traits was that the mutant number of nodes on main stem was less 6 than control group of average nodes on main stem of 11; the average of mutant plant height of 81.1 cm, was significantly lower than the control group (160.5cm); the mutants average weight of spike of 1.65 g, was significantly lower than control group(20.67g);the mutant heading time was earlier 28 days than the average of control group heading date.The result showed that there existed significant variation between normal and mutant strain in the main agronomic traits.For identifying its origination,we used SSR molecular marker method to identify it from the natural mutation and without mixing the hybrid seed.We also tested early heading plants identified in 2012 by EMS mutagenesis of Jingu 21and 420 millet germplasm resources for early flowering date.In total,13 materials out of 40 EMS mutagenesis of Jingu 21 were screened,219 varieties out of 420 millet germplasm resources showed eraly flowering compared with Yugu 1.We also analyzed the result of genomic resequencing of Jingu 21 compared with Yugu 1.The depth coverage of genome was about 34×.The coverage of genome added up to 95.34%.The raw reads were up to 171.63M,15.45GFinally,we obtained 166.1M clean reads and clean 14.95Gbp base;the total number of SNP genome was for 1,167,555,small fragment of InDels were 169,805,the total number of structural variation was 54,297; the number of variation of genes was 16,357, the number of notation to the COG database of the UniGene was 9,206, the number of notation to UniGene GO database was 22,081, the number of notation to the KEGG database was 6,375, the number of notation to the Swissprot database was 19,520, the number of notation to the NR database was 28,621.From SNP, InDel and SV, the specific sequence could be used to explore the excellent Jingu 21 characteristic of early flowering mutant gene and provide abundant molecular marker data.
