A Comparative Study On Reproductive Characteristics Of Different Paulownia Clones

Paulownia is Scrophulariaceae(Scrophulariaceae) Paulownia, Paulownia species collectively, is a fast-growing timber species, economic value in industry, agriculture,medicine, national defense, gardens, and widely used in daily life, and high value.Jiangxi academy of forestry breeding in 2014 and plans to declare provincial forests for five paulownia fine clones ‘C22’, ‘C23’, ‘Za Wu-12’, ‘Bai Dan-25’, ‘Huang Jintong’, asexual reproduction such as adopting traditional thong, grafting method, article root and grain supply ability is limited, difficult to get a lot of high quality nursery stock in the short term,which prevented breeding achievements into productivity as quickly as possible.So we must explore new breeding way, to the existing production capacity, as soon as possible to establish a new efficient breeding system.This study was based on the above reasons, plans to build five paulownia clones seed efficient breeding system, groped in its breeding conditions, on the basis of improving paulownia somaclone of large-scale production capacity.The main conclusions are as follows:1. 5 paulownia fine clones of culture conditions such as ‘C22’ no difference, the best temperature can choose(25 ± 2) ℃, the best training intensity of 2000 lx, illumination time of 12 h/d.2. For the 5 Paulownia Clones such as‘C22’, the best synthesis method for establishing the aseptic ramets was: taken the middle root coppice shoot as explants, using0.1% mercury to asepticize the explants for four minutes, with the cultural medium(MS+6-BA0.5mg/L(the unit as same as follows) + IBA0.1) for buds inducing.3. ‘C22’, ‘C23’ and ‘Huang Jintong’ optimum proliferation medium for MS + 6-BA4.0 + IBA0.1, differentiation rate above 95%, and multiplication factor above 6.0;‘Za Wu-12’ and ‘Bai Dan-25’ one of the most appropriate proliferation medium for MS +6- BA5.0 + NAA0.2, differentiation rate was 95%, and the highest multiplication factor above 5.5. The proliferation and subculture of subculture was appropriate for 25 d.4. ‘C22’, ‘C23’ and ‘Huang Jintong’ optimal rooting medium for MS + IBA0.1,rooting rate was 100%; ‘Za Wu-12’ and ‘Bai Dan-25’ rooting medium to MS + NAA0.2 is the best. The best times for transplant is from April to June and September to November,Field transplant, the survival rate of over 70%.then transplanted into the matrix for the rooting rate can reach 80%.
