In this study, histone deacetylase HDA903 gene from 84K populus (Populus alba X Populus gllandulosa) was cloned. The sequence and expression of HDA903 under drought stress condition were analyzed. The drought tolerance of HDA903 transgenic tobacco was determined. The full length of HDA903 ORF is 1503 bp, encoding a protein composed of 500 amino acids. The HDA903 is a hydrophilic acidic protein with anisoelectric point of 5.26. HDA903 protein contains a conserved domain, HDAC domain, and has close relationship with HDA903 from Populus trichocarpa. The expression levels of HDA903 in organs were analysed using real-time PCR. The results showed that the transcripts of HDA903 gene were detected in leaves, stems and roots and the mRNA levels were different in three organs. The expression of HDA903 was regulated by drought. Under drought stress condition, the expression of HDA903 was down-regulated. After drought treatment for 12h or 24h, the expression level of HDA903 reached the lowest level and then increased steadyly.The expression patterns of HDA903 gene in leaves, stems and roots were same.The expression of 84KHDA903 in tobacco rendered the transgenic plants high tolerance to drought stress. In addition, expression of 84KHDA903 reduced the seed dormancy, thus increased germination of the seeds.
