Rice blast, which is caused by the ascomycotina fungus Magnaporthe oryzae, is one of the most serious disease that damage to the rice on large scale all over the world. To breed a rice variety with resistance to the rice blast is the most economical and effective way to control the disease. Two main effect gene, namely Pi47and Pi48, was mapped on the chromosome11and12, respectively, in the indica rice cultivar Xiangzi3150(XZ3150), which exhibiting durable resistance to rice blast for more than30years in Hunan Province, China in the previously studies. Fine mapping and cloning the Pi48gene were conducted in this study. Meanwhile, a series of near-isogenic lines (NILs), which with the CO39genetic background, introgressed one of some rice blast resistant gene that fine mapped in our team.The main results are as following.1. A series of near-isogenic lines (NILs), which with the susceptibility variety CO39as the recurrent parent, and recombinant inbred lines (RIL), which harboured Pi47, Pi48gene derived from the cross of CO39/XiangZi3150, Pi49gene from CO39/mowanggu, and Pi2-1gene from CO39/Tianjinyeshengdao, respectively, as the donor parents, were developed by continuous backcross with molecular markers assisted selection.2. The Pi48gene was fine mapped between the maker interval of RM27933-LY48, with the BC4F2population derived from the cross of the near isogenic line and the recurrent parent.3. A monoclonal that contains interested fragments was screened from the BAC library of the resistant rice variety of Xiangzi3150, by using the PCR method with the mixed BAC as template and with the markers of LY48, LY51and LY56as primers, respectively.4. Bioinformatics analysis showed that two chromosome regions coding NBS-LRR class structure were existed in the chromosome interval where the Pi48gene located. The sequence of one candidate gene coding a gene similar to the cloned resistance gene of Pita, whereas, the other one coded a not reported functional gene.
