The objective of this study was to research sodium fluorid’s reproductive toxicity on adult zebrafish, to provide theoretical basis and experience for the further research of fluorid’s reproductive toxicity on fish (aquaculture). We choosed zebrafish as the experimental animal in this subject research, and sodium fluoride was the experimental compound in the chronic toxicology test. Firstly, we should try to determine the median lethal concentration of sodium fluoride for adult zebrafish by acute toxicity test which will provides necessary basis and scientific reference for settings of fluorine concentration gradient in chronic toxicity studies; And then began to do the chronic toxicology test, experiments had to be done respectively after exposured to sodium fluoride for 45 d and 90d :To detect the quantity of the expression of related genes and hormone level changes in the zebrafish through enzyme-linked immunoassay (ELISA) and real-time fluorescent quantitative PCR (QRT-PCR), and to observe the development of zebrafish gonad(testis and ovaries) through ordinary HE dyeing technology.The result shows that:Inside the female adult zebrafish:(1) The content of estradiol (E2)/vitellogenin (VTG) reduced, and the content of testosterone (T) improved after exposured to sodium fluoride by enzyme-linked immunoassay (ELISA); (2) It can be found that the relative expression of progesterone receptors (Prg) showed a trend of droped in the first time then rised with the increase of the fluoride concentration (the expression in 40 mg/L group decreased but it increased in 80 mg/L group)/the relative expression of glucocorticoid receptor (GR) showed a same trend with progesterone receptors (Prg) with the increase of fluorine concentration (the expression in 40 mg/L group decreased but it increased in 80 mg/L group)/the relative expression of androgen receptor (AR) rised with the increase of fluorine concentration/the relative expression of CYP19a showed a trend of overall declined after exposured to sodium fluoride by real-time fluorescent quantitative PCR detection (QRT-PCR); (3) We can found that the ovarian development is restricted/follicular dysplasia appears after exposured to sodium fluoride by tissue section general HE staining.In the male adult fish:(1) The content of estradiol (E2)/vitellogenin (VTG) was enhanced, and the content of testosterone (T) reduced after exposured to sodium fluoride by enzyme-linked immunoassay (ELISA); (2) It can be found that the relative expression of progesterone receptors (Prg) showed a trend of rised in the first time then droped with the increase of the fluoride concentration (the expression in 40 mg/L group increased but it decreased in 80 mg/L group)/the relative expression of glucocorticoid receptor (GR) showed a trend of rised in the first time then droped with the infected time delay (the expression increased after 45 d but it decreased after 90 d)/the relative expression of androgen receptor (AR) declined with the increase of fluorine concentration/the relative expression of CYP19a showed a overall upward trend after exposured to sodium fluoride by real-time fluorescent quantitative PCR detection (QRT-PCR); (3) We can found that the sperm production capacity of testis reduced/testis growth was stunted after exposured to sodium fluoride by tissue section general HE staining.The test results show that sodium fluoride can influence the genes expression quantity related to reproduction/hormones level/gonad development and produce toxic effect to zebrafish reproduction eventually.
