Bagging is one of the usual technique of the cultivation and Management in fruit trees. Fructose, glucose and sucrose are the major accumulation, The unloading and transport of photoassimilate will be affected by micro-environment in bag and the internal quality of apple fruit will be influenced directly. Hexokinase is one of the important enzyme for the pathway of sugar accumulated, meanwhile is close to the plant respiratory metabolism,so is crucial to control the sink strength and related to development of apple fruit closely. The effects of the accumulation and metabolism of hexose, the activity of the hexokinase, the localization of the hexokinase and the content of hexokinasel in pericarp and core flesh tissues were studied in ’Malus domestica Borkh.cv.FujiNo.2’ apple fruit bagged by plastic film bag and paper bag. The results showed that:1. In different stages of apple fruit, the protein of hexokinasel increased gradually, HXK1 localized in sepal vascular was more than pericarp vascular. HXK1 was localized in vascular bundle of sepal and pericarp except vessel and surrounding vascular bundle sheath cell;2. In different stages of apple fruit, the hexokinasel protein was significantly more than it’s in apple fruit bagged by plastic film bag, paper bag and control, the hexokinasel protein was significantly more than it’s in the earlier stage in apple fruit bagged by plastic film and control, but it’s not in the later stage, HXK activity was decline in core and pericarp flesh tissue. HXK activity of bagging apple fruit with paper bag was great significant higher than fruit bagged by plastic bag and control;3. In apple fruit of different stages, the content of glucose in core flesh tissue in fruit bagged by paper was significant higher than fruit bagged by plastic and control from 85 days to 125 days. On the 125 days after full bloom, the content of sucrose in flesh tissue of fruit bagged by plastic and control was significant higher than fruit bagged by paper bag, and the content of sucrose in apple flesh tissue reached a rapid growth in bagging and control fruit.The content of fructose of fruit bagged by paper bag was significant lower than fruit bagged by plastic and control, at the same time,the content of fructose was lower than every other period in bagging and control fruit;4. The apple fruit weight was obviously influenced by the paper bag, the fruit weight of bagged by plastic and control was significant higher than bagged by paper bag, compare fruit bagged by plastic with control and bagged by paper, the assimilation substance accumulated earlier.
