| Fusarium oxysporum f.sp.niveum.FON mumant strain that has resistence to marigold spionsad has been obtained by the use of chemical mutagenic method under a selection pressure of magrigold spinosad.By advantage of prior achievement,the transcriptome sequencings of both wild-type strain of FON and mutant strain have been done. All unigenes number is 23,042 and there are 827 differentially expressed unigenes.Two expressed unigenes Locus18025 and Locus3324 are annotated as fucose permease and express highly remarkable highly in the mutant strain than wild-type. Futhermore,cloning of their CDS and spatial structure predition of their protein products have been done. And, we design different medias using different sugars as carbon source to research the variety of the capacity to transport and use different kinds of sugar between two type strains.By cloning and bioinformatic methods, Locus18025 and Locus3324 are identified as 1-fucose premease,named fucP1 and fucP2. And,there is no nucleic acid sequence mutant in two genes CDS in the mutant strain.The result of culture experiment show that both colony morphology and gowth speed are different between the two FON strains. The varitety of mutant strain colony morphology indicates nutritional approach has changed in the mutant strain of FON.The wild-type strain grows speedly on the galactose media and starch media,but slowly in 1-fucose media comparatively. The mutant strain grows speedly on the 1-fucose media and starch media,but slowly in galactose media comparatively.So we can draw a conclusion,that,the mutant strain of FON, which upregulated the expression of fucPl and fucP2, significantly alter the utilization of fucose as carbon resource. In the same words,due to gene expression upregulation of fucPl and fucP2, the cells of the mutant strain can transport more fucose intracellular and use it,ultimately the increased utilization of fucose make it grow faster than wild-type strain.Furthermore, for the mutant strain of FON,altering the carbon utilization pathway including upregulation of the capalility of using fucose in one of its anti-marigold spinosad mechanism. That is one of spinosad marigold anti-FON mechanism is to inhibit carbon utilization of Fusarium oxysporum. |
