Sex differentation and determination is a complex bioliogiacal process which composed of many interacting events in fish, females and males will develop different sex in the same genetic material and environment factors.Yellow catfish(Pelteobagrus fulvidraco) has been a freshwater aquaculture species in China. It is attracted much attention for commercial interest.In this study, Gonadal differentiation and development of 3-60 day-old yellow catfish Pelteobagrus fulvidraco reared artificially were observed by histological techniques in the study. The results showed that differentiation was found to be earlier in the ovary than that in the testicle at water temperature of(20 ± 4)℃, yellow catfish are a gonochoristic differentiation. The ovarian differentiation was found to be began 14 days after hatching, and showed the formation of organic heave(OH), gradually forming the ovarian cavity. However, the testicular differentiation was found to be occurred 54 days after hatching, and to show the formation of spermaduct anlage(SA), gradually forming the seminiferous duct. The yellow catfish transcriptomes were sequenced with high throught Illumina sequencing, The samples were selected according to the time of gonadal differentiation, the beginning time(3d after hatching, called B) is unkown sexes and recognized as control, B was blended by using three fish. The ovarian differentiation was found to be began at 14 d after hatching and the testicular differentiation was found to be occurred at 54 d after hatching, female and male were visually identified at 90 d after hatching, analyzed the gonad transcriptome and found sex-related genes of yellow catfish. Finally sex-related gene expressions were found and validated by using quantitative real-time PCR(qRT-PCR).The resulting data and transcriptome sequencing genomes were compared, the results show a total of expression get 203,660,437 Raw reads from 9 transcriptomes in male and female gonads, Clean reads get filtered as 191,297,677. Reads quality control carried FPKM with calculation, obtaining FPKM values of all genes, the result for the gene expression values with a larger difference, the results are displayed as a demarcation point B and 14, 15, 54, Y and 55 are male in the front of B, female whose FPKM relatively low value; females are 13 and 53 behind B respectively. Unknown gender catfish were identified at the transcriptional level. With the nr database for comparison, the results show that five of catfish found gender-related differences in male and female catfish expressed genes, these genes are the same genes ZP3(zona like sperm-binding protein 3). By qRT-PCR for transcriptome sequencing results validate the findings of variation ZP3 gene expression showed that the expression trends are similar results with transcriptome sequencing.
